摘要
以非洲紫罗兰为试验材料,以其无菌苗叶片为外植体,建立了愈伤组织诱导及植株再生体系。试验结果表明:MS+6-BA2.0mgL+NAA0.5mgL培养基配方诱导愈伤组织效果最好,出愈率可达73.9%,暗培养不利于愈伤组织诱导;在培养基MS+6-BA1.0mgL+NAA0.1mgL中愈伤组织分化不定芽的分化系数最高可达14.0;MS+NAA0.5mgL+IAA0.5mgL培养基可使生根率达到100%,平均单株生根数达12.4。
Callus induction and plant regeneration system were constructed by using leaves of sterile Saintpaulia ionantha as explants.Results showed callus induction ratio were 73.9% and induction efficiency was best in the medium MS+6-BA 2.0mgL+NAA 0.5mgL.Darkness is not conducive to callus induction.Differentiation coefficient of callus was highest up to 14.0 in the medium MS+6-BA 1.0mgL+NAA 0.1mgL.Rooting percentage was up to 100% in MS+NAA 0.5mgL+IAA 0.5mgL medium,the average roots of each plant were 12.4.
