通腑化瘀清胰汤对急性胰腺炎小鼠肠道菌群影响的研究

摘要 目的探讨通腑化瘀清胰汤对急性胰腺炎(AP)小鼠肠道菌群的影响。方法将50只成年且无特定病原体C57BL/6小鼠按随机数字表法分为空白对照组、AP模型组、通腑化瘀清胰汤组、奥曲肽组及大承气汤组,每组10只;除空白对照组外,其余4组均采用小鼠腹腔注射雨蛙素法构建AP模型。造模成功后,通腑化瘀清胰汤组、大承气汤组分别按23.5 mL/(kg·d)灌胃通腑化瘀清胰汤、大承气汤,奥曲肽组按10μg/(kg·d)背部皮下注射奥曲肽注射液,空白对照组、AP模型组仅予观察处理,连续给药3 d后采用颈椎脱臼法处死小鼠。取结肠组织进行HE染色;采集盲肠内新鲜粪便进行16S rDNA高通量测序和生物信息学分析,比较5组小鼠肠道菌群多样性和优势菌属。结果与空白对照组比较,AP模型组结肠组织出现大量炎性细胞浸润,隐窝缺失,组织损伤严重,Chiu评分明显升高(P<0.05),Shannon、Chao1、Simpson指数均明显降低(均P<0.05);与AP模型组比较,通腑化瘀清胰汤组小鼠结肠组织损伤和炎性细胞浸润情况均有所好转,Chiu评分均明显降低(均P<0.05),Chao1、Shannon、Simpson指数均明显升高(均P<0.05)。β多样性主坐标分析显示,通腑化瘀清胰汤组与AP模型组样本之间距离较远,空白对照组与AP模型组样本之间距离较远,空白对照组与通腑化瘀清胰汤组、奥曲肽组、大承气汤组样本之间距离较近。与AP模型组比较,通腑化瘀清胰汤组、空白对照组乳杆菌属丰富度均明显减少(均P<0.05),而瘤胃球菌属、颤螺菌属丰富度均明显增加(均P<0.05)。物种差异分析显示,空白对照组区别于其他4组的优势菌属主要为别样棒菌门、双歧杆菌属;AP模型组区别于其他4组的优势菌属主要为乳杆菌、假单胞菌;通腑化瘀清胰汤组区别于其他4组的优势菌属主要为消化球菌科、颤螺菌属;大承气汤组区别于其他4组的优势菌属主要为韦荣球菌科、脱铁杆菌科 Objective To investigate the effect of Tongfu-Huayu-Qingyi(THQ)decoction on intestinal flora of acute pancreatitis(AP)mice.Methods Fifty adult specific-pathogen-free C57BL/6 mice were divided into blank control group,AP model group,THQ group,octreotide group and Dachengqi group,with 10 mice in each group.Except for the blank control group,the AP model was induced by intraperitoneal injection of hyranolin in mice of other 4 groups.After successful modeling,THQ group and Dachengqi group received THQ decoction or Dachengqi decoction 23.5 mL/(kg·d)by gavage,respectively;while octreotide group received subcutaneous injection of octreotide 10μg/(kg·d).The mice were sacrificed after continuous administration for 3 days.Colon tissue was taken for HE staining.Fresh fecal samples were collected from cecum,16S rDNA high-throughput sequencing and bioinformatics were used to analyze the diversity and difference of intestinal flora in 5 groups of mice.Results Compared with blank control group,colon tissue in AP model group showed a large number of inflammatory cell infiltration,absence of crypts and serious tissue damage,the HE score was significantly increased(P<0.05),Shannon index,Chao1 index and Simpson index were significantly decreased(both P<0.05).Compared with AP model group,colon tissue injury and inflammatory cell infiltration of mice in THQ group were alleviated,HE score was significantly decreased(all P<0.05),Chao1 index,Shannon index and Simpson index were significantly increased(all P<0.05).The principal coordinate analysis of β diversity showed that the distance between THQ group and AP model group was far,the distance between blank control group and AP model group was far,and the distance between blank control group and THQ group,octreotide group and Dachengqi group was close.At the genus level,the abundance of Lactobacillus in AP model group was higher than that in THQ group and blank control group(all P<0.05),while the abundance of Ruminococcus and Tremillum decreased(all P<0.05).The species difference ana
