摘要
目的分析广东河源地区叶酸代谢相关酶亚甲基四氢叶酸还原酶(MTHFR)及甲硫氨酸合成酶还原酶(MTRR)基因位点多态性与妊娠期高血压疾病的关系。方法采用回顾性分析方法,选取2022年1月至2022年12月孕期在河源市妇幼保健院行叶酸代谢相关酶基因位点(MTHFR基因C677T、A1298C位点以及MTRR基因A66G位点)检测且孕晚期在本院产科住院分娩的孕妇作为研究对象,将100例合并有妊娠期高血压疾病的孕妇纳入实验组,200名无妊娠期高血压疾病的健康孕妇纳入对照组,比较两组两个基因三个位点的多态性情况。结果实验组MTHFR C677T位点TT基因型频率及T等位基因频率高于对照组,差异有统计学意义(P<0.05);实验组MTHFR A1298C位点AA/AC/CC三种基因型频率以及A、C等位基因频率,MTRR A66G位点AA/AG/GG三种基因型频率以及A、G等位基因频率,与对照组相比,差异无统计学意义(P>0.05)。结论河源地区MTHFR基因C677T位点多态性与妊娠期高血压疾病有关,可以通过检测孕妇MTHFR基因C677T位点为临床上防治妊娠期高血压疾病以及个性化叶酸的增补提供理论指导。
Objective To analyze the relationship between polymorphisms in the gene loci of methylenetetrahydrofolate reductase(MTHFR)and methionine synthase reductase(MTRR)related to folate metabolism in the Heyuan region of Guangdong province and gestational hypertension.Methods A retrospective analysis method was used to select pregnant women who underwent folate metabolism related enzyme gene loci(MTHFR gene C677T,A1298C locus,and MTRR gene A66G locus)detection at Heyuan Maternal and Child Health Hospital from January 2022 to December 2022.Subsequently,pregnant women who delivered in our hospital's obstetrics department in the late pregnancy were selected as the research subjects.100 pregnant women with gestational hypertension were included in the experimental group,200 healthy pregnant women without gestational hypertension were included in the control group to compare the polymorphism of two genes and three loci between the two groups.Results The TT genotype frequency and T allele frequency at the MTHFR C677T locus in the experimental group were higher than those in the control group,with statistical significance(P<0.05).The experimental group showed no statistically significant differences in the frequencies of the three genotypes AA/AC/CC and A,C alleles at the MTHFR A1298C locus,AA/AG/GG at the MTRR A66G locus,and A,G alleles compared to the control group(P>0.05).Conclusion The C677T polymorphism of MTHFR gene in Heyuan region is related to gestational hypertension.Detecting the C677T locus of MTHFR gene in pregnant women can provide theoretical guidance for clinical prevention and treatment of gestational hypertension and personalized supplementation of folic acid.
