摘要
目的研究沧州地区5,10-亚甲基四氢叶酸还原酶(MTHFR)基因多态性与妊娠期高血压疾病的相关性。方法选取2017年1月1日至2017年12月31日期间来沧州市妇幼保健院就诊的50例妊娠期高血压患者作为实验组对象,另选取同时期无妊娠高血压的50例孕妇作为对照组对象,使用荧光定量PCR技术对两组MTHFCR基因C667T及A1298C位点的多态性进行测定。结果试验组与对照组的MTHFR基因C667T位点的C/C型和C/T型的基因频率之间均无统计学差异(χ2=1.967,0.794,均P>0.05)。试验组T/T型相较于对照组,基因型频率明显高于后者,差异有统计学意义(χ2=6.453,P=0.011)。试验组与对照组之间等位基因频率有明显的统计学差异(χ2=6.810,P=0.009),试验组等位基因T的频率明显高于对照组。两组间的MTHFR基因A1298C位点的多态性无统计学差异。结论沧州地区MTHFR基因C667T位点的多态性与妊娠期高血压的发生有密切相关性,而A1298C位点与妊娠期高血压无明显相关性。临床上可采取MTHFR基因C667T位点多态性筛查来评价妊娠期高血压的预后情况。
Objective:To study the association of 5,10-methylenetetrahydrofolate reductase(MTHFR)gene polymorphisms with pregnancy-induced hypertension in Cangzhou. Methods:Fifty hypertensive patients with pregnancy during the period from January 1,2017 to December 31,2017 in Cangzhou Maternal and Child Health Hospital were selected as experimental subjects. Fifty pregnant women without gestational hypertension at the same time were selected as the experimental group. In the control group,the polymorphisms of the C667 T and A1298 C loci of the two MTHFR genes were determined using real-time PCR. Results:There was no significant difference between C/C type and C/T type gene frequency of MTHFR gene C667 T in the experimental group and the control group(χ2=1.967,0.794,all P>0.05). Compared with the control group,the frequency of genotypes in the T/T group was significantly higher than that of the latter group(χ2=6.453,P=0.011). There was a significant difference in allele frequencies between the experimental group and the control group(χ2=6.810,P=0.009). The frequency of allele T in the experimental group was significantly higher than that in the control group. There was no statistical difference in the polymorphism of the A1298 C locus of MTHFR gene between the two groups. Conclusion:The polymorphism of the MTHFR gene C667 T locus in Cangzhou region is closely related to the occurrence of pregnancy-induced hypertension. There is no significant correlation between the A1298 C locus and pregnancy-induced hypertension. MTHFR gene C667 T polymorphism screening can be used to evaluate the prognosis of pregnancy-induced hypertension.
