摘要
目的制备含有苄达赖氨酸和透明质酸的角膜接触镜,研究透明质酸对苄达赖氨酸释放行为的影响。方法采用紫外引发聚合法制备水凝胶角膜接触镜,以透光率、含水量和离子渗透率为指标筛选最佳聚合单体溶液配比和紫外固化时间;利用紫外分光光度法建立了苄达赖氨酸体外分析方法;采用聚合单体—药物共聚法和吸附法制备两种载有苄达赖氨酸的角膜接触镜,并比较了两种载药角膜接触镜体外药物释放行为;在载有苄达赖氨酸的角膜接触镜中加入透明质酸,考察了透明质酸对苄达赖氨酸释放的调控效果。结果水凝胶角膜接触镜聚合单体溶液配比为HEMA用量0.863 g,EGDMA用量0.043 g,Photointiator 1173用量0.015 g,紫外固化时间为6 min,可形成最佳效果水凝胶角膜接触镜。水凝胶角膜接触镜在可见光650 nm处测得透光率为(94.3±0.4)%、含水量为(57.6±0.5)%、离子渗透率为(2.06×10^(-6)±0.06)cm^(2)·s^(-1),符合国家标准。聚合单体—药物共聚法制备的角膜接触镜中苄达赖氨酸具有更长的缓释时间,96 h的累计释放率为(91.59±3)%;吸附法制备的苄达赖氨酸角膜接触镜中,苄达赖氨酸48 h内几乎全部释放。聚合单体—苄达赖氨酸—透明质酸(HA)共聚法制备的角膜接触镜可进一步延缓苄达赖氨酸的释放。当HA浓度为1 mg·mL^(-1)时,苄达赖氨酸96 h的累积释放率为(68.89±5)%;当HA浓度为1.5 mg·mL^(-1)时,苄达赖氨酸96 h的累积释放率为(55.06±4)%;当HA浓度为2 mg·mL^(-1)时,苄达赖氨酸96 h的累积释放率为(50.03±2)%。结论聚合单体—药物—透明质酸共聚法制备的角膜接触镜具有缓慢释放的作用。
Objective To prepare contact lenses containing bendazac lysine and hyaluronic acid,and to study the effect of hyaluronic acid on the release behavior of bendazac lysine.Methods The hydrogel contact lens was prepared by UV initiated polymerization,and the best ratio of monomer solution and UV curing time were selected according to the transmittance,water content and ion permeability;A method for in vitro analysis of bendazac lysine was established by UV spectrophotometry;Two kinds of drug loaded corneal contact lenses were prepared by polymeric monomer drug copolymerization and adsorption methods,and their drug release behaviors in vitro were compared;Hyaluronic acid was added to the contact lens containing bendazac lysine to investigate the effect of hyaluronic acid on the release of bendazac lysine.Results The best effect of hydrogel contact lens could be obtained when the ratio of polymer monomer solution of hydrogel contact lens was 0.863 g HEMA,0.043 g EGDMA,0.015 g Photointiator 1173,and UV curing time was 6 min.The transmittance,water content and ion permeability of the hydrogel contact lens measured at 650 nm of visible light were(94.3±0.4)%,(57.6±0.5)%and(2.06×10^(-6)±0.06)cm^(2)·s^(-1),which were in accordance with national standards.In the corneal contact lens prepared by the polymerization monomer drug copolymerization method,bendazac lysine had a longer sustained release time,and the drug release rate was(91.59±3)%at 96 h.In the bendazac lysine corneal contact lens prepared by the adsorption method,bendazac lysine could realize complete release within 48 h.The contact lens prepared by copolymerization of polymer monomer bendazac lysine hyaluronic acid(HA)can further delay the release of bendazac lysine.When the concentration of HA was 1 mg·mL^(-1),the cumulative release rate of bendazac lysine for 96 h was(68.89±5)%;When the concentration of HA was 1.5 mg·mL^(-1),the cumulative release rate of bendazac lysine for 96 h was(55.06±4)%;When the concentration of HA was 2 mg·mL^(-1),the cumulativ
