摘要
目的探究黄连解毒汤对逆转产金属酶铜绿假单胞菌(PA)耐药性的能力。方法通过纸片扩散法和PCR法筛选出产金属酶PA后,通过R质粒接合试验筛选出存在R质粒的菌株;二倍稀释法确定筛选出的试验菌株对黄连解毒汤的最低抑菌浓度,以亚抑菌浓度进行R质粒消除试验,通过平板影印法验证消除子,计算消除率。结果11株blaIMP基因型为阳性的PA中进行质粒接合转移试验,有3株(27.27%,3/11)R质粒接合试验阳性;黄连解毒汤和5%十二烷基硫酸钠(Sodium dodecyl sulfate,SDS)对3株菌株在24h、48h和72h内质粒平均消除率分别为0%、0.78%、1.56%以及0.11%、2.44%、1.89%。结论黄连解毒汤可以通过消除R质粒逆转产金属酶PA的耐药性,但逆转率低于金属酶化学抑制剂SDS。
Objective To investigate the ability of Huanglian Jiedu decoction to reverse the drug resistance of metalloenzyme-producing Pseudomonas aeruginosa.Methods After screening Pseudomonas aeruginosa producing metalloenzyme using paper diffusion and PCR method,the strains with R plasmid were screened through R plasmid conjugation test.The minimum inhibitory concentration of the selected experimental strains on Huanglian Jiedu decoction was determined by double dilution method.The R plasmid elimination test was conducted at subinhibitory concentration,and the elimination rate was calculated by verifying the eliminators using plate photocopying method.Results Among the 11 PA strains with positive bla IMP genotype,plasmid conjugation transfer test were conducted,and 3 strains(27.27%,3/11)showed positive R plasmid conjugation test.The average plasmid elimination rates of Huanglian Jiedu decoction and 5%Sodium dodecyl sulfatel(SDS)on 3 strains were 0%,0.78%,1.56%and 0.11%,2.44%,1.89%respectively within 24h,48h and 72h.Conclusion Huanglian Jiedu decoction can reverse the drug resistance of metalloenzyme-producing Pseudomonas aeruginosa by eliminating R plasmid.The reverse rate is lower than that of metalloenzyme chemical inhibitor SDS.
