摘要
从林下山参根际土壤中筛选到具有溶磷作用的根际细菌,明确其对人参生长的促生作用和机制。利用平板稀释法对根际细菌进行分离和筛选;利用16S rRNA进行鉴定;通过功能培养基检测菌株溶磷能力;室内盆栽试验采用灌根法将溶磷菌接种于人参根际,培养90 d后测定人参生理指标及根际土壤理化性状。结果表明,从林下山参根际土壤中共获得3株溶磷菌,经鉴定分别为Nocardioides allogilvus(FT4)、Pseudomonas reactans(FT6)和Bacillus tequilensis(FT10),其中FT6溶磷效果最好,该菌株能同时溶解无机磷和有机磷,溶磷量分别为390.99 mg/L和11.53 mg/L。与对照相比,FT6使人参地上部鲜重和干重分别提高45.8%和35.0%;地上部磷含量和磷吸收率分别提高106.2%和178.0%;人参叶片PSⅡ最大光合效率和实际光合效率分别提高32.3%和55.2%(P<0.05);FT6能使土壤pH降低0.45,土壤速效磷含量、酸性磷酸酶活性和植酸酶活性分别提高11.2%、42.5%和595.5%(P<0.05)。本研究表明,FT6具有较强的溶磷作用,能促进人参生长,并具有改良人参栽培土壤的潜力,在人参栽培产业的绿色可持续发展中具有较高的应用价值。
Screening rhizosphere bacteria with phosphorus-solubilizing ability fromthe rhizosphere soil of ginseng that growing in the forest,and clarifying their promoting effect and mechanismon ginseng growth.Aplate dilution method was used to isolate and screen the rhizosphere bacteria which were then identified by 16S rRNA sequencing.The functional mediumwas used to detect the phosphorus solubilization ability of the strains.Indoor potted experiment was conducted,and phosphorus solubilizing bacteria were inoculated into the 1-year-old ginseng rhizosphere through root irrigation.Both the physiological indexes of ginseng and physical and chemical properties of ginseng rhizosphere soil were measured after 90 days.Three phosphate solubilizing bacteria strains were obtained from the ginseng rhizosphere soil,naming Nocardioides allogilvus(FT4),Pseudomonas reactans(FT6),and Bacillus tequilensis(FT10),among them,FT6 showed the highest phosphorus-solubilizing ability;and was able to dissolve both inorganic and organic phosphorus,with a dissolved phosphorus amount of 390.99 mg/L and 11.53 mg/L,respectively.Compared to the control,FT6 increased fresh weight and dry weight of the aboveground of ginseng by 45.8%and 35.0%,respectively,increased the phosphorus content and phosphorus absorption efficiency of ginseng by 106.2%and 178.0%,respectively,and increased the PSⅡmaximum photosynthetic efficiency and actual photosynthetic efficiency of ginseng leaves by 32.3%and 55.2%,respectively(P<0.05).In addition,FT6 decreased the rhizosphere soil pH by 0.45 and increased soil available phosphorus content,acid phosphatase activity and phytase activity by 11.2%,42.5%and 595.5%,respectively(P<0.05).These findings suggested that the bacteria FT6 had strong phosphate-solubilizing ability,and could promote ginseng growth and had the potential to improve the quality of soil that cultivating ginseng,which indicating its strong application value in the green and sustainable development of ginseng cultivation industry.
