摘要
目的 探讨氯米芬联合补肾种子汤调控子宫内膜白血病抑制因子(Leukemia inhibitory fa-ctor,LIF)表达水平,研究其影响多囊卵巢综合征(Polycysticovarysyndrome,PCOS)雌鼠子宫内膜容受性的相关机制。方法 将44只8周龄SPF级SD雌性大鼠采用随机数字表法分为对照组10只、造模组34只(造模成功后,对照组和造模组大鼠各随机处死2只),造模组SD雌鼠颈背部皮下注射丙酸睾酮+绒毛膜促性腺激素(Human chorionic gonadotropin,HCG)构建PCOS动物模型。将造模组剩余32只SD雌鼠采用随机数字表法分为模型组、氯米芬组、补肾种子汤组、氯米芬联合补肾种子汤组,每组各8只,分别给予生理盐水、氯米芬、补肾种子汤、氯米芬联合补肾种子汤药物干预,连续14 d,绘制大鼠体质量增长变化曲线。大鼠处死后,采用Elisa法检测血清性激素黄体生成素(Luteinizinghormone,LH)、促卵泡素(Follical stimulating hormone,FSH)、睾酮(Testosterone,T)含量,测量子宫内膜厚度,行子宫HE染色,免疫组化观察LIF在子宫内膜组织中表达水平。结果 药物干预后与模型组比较,氯米芬组FSH值升高,T值降低,LH、LH/FSH值降低,差异有统计学意义(P<0.05,P<0.01);补肾种子汤组LH、T值降低,FSH值升高,LH/FSH值降低,差异有统计学意义(P<0.05,P<0.01);氯米芬联合补肾种子汤组LH、LH/FSH、T值降低,FSH值升高,差异有统计学意义(P<0.01)。与氯米芬组比较,氯米芬联合补肾种子汤组FSH值升高,T值降低,差异有统计学意义(P<0.05)。与补肾种子汤组比较,氯米芬联合补肾种子汤组FSH值升高,LH/FSH、LH、T值降低,差异有统计学意义(P<0.05,P<0.01)。药物干预后氯米芬组、补肾种子汤组、氯米芬联合补肾种子汤组子宫内膜仍存在内膜层菲薄,与对照组比较,差异有统计学意义(P<0.05),表明药物干预疗程不足;与模型组比较,氯米芬组、氯米芬联合补肾种子汤组子宫内膜变厚,差异有统计学意义(P<0.05)
Objective To investigate the regulatory effect of clomiphene combined with Bushen Zhongzi Decoction on the expression of leukemia inhibitory factor(LIF)in the endometrium and explore its mechanism on endometrial receptivity in female rats with polycystic ovary syndrome(PCOS).Methods Forty-four 8-week-old female SD rats were divided into a control group(n=10)and an experimental group(n=34)using a random number table,with two rats randomly euthanized from each group after successful modeling.The PCOS model was induced in the experimental group by subcutaneous injection of testosterone propionate and human chorionic gonadotropin(HCG)in the neck and back.The remaining 32 model rats in the experimental group were further divided into a model group,a clomiphene group,a Bushen Zhongzi Decoction group,and a combination group(clomiphene + Bushen Zhongzi Decoction)according to a random number table,with eight rats in each group.The rats were given physiological saline,clomiphene,Bushen Zhongzi Decoction,or the combination of clomiphene and Bushen Zhongzi Decoction for 14 days.The body weight growth curve of rats was plotted.After euthanasia,serum levels of luteinizing hormone(LH),follicle-stimulating hormone(FSH),and testosterone(T)were measured using ELISA.Endometrial thickness was measured and the uterus underwent HE staining.Immunohistochemistry(IHC)was used to observe LIF expression levels in the endometrial tissue.Results Compared with the model group,the clomiphene group showed increased FSH levels,decreased T levels,and decreased LH and LH/FSH ratios(P<0.05,P<0.01).The Bushen Zhongzi Decoction showed decreased LH and T levels,increased FSH levels,and decreased LH/FSH ratios(P<0.05,P<0.01).In the combination group,LH,LH/FSH,and T levels decreased,and FSH levels increased significantly(P<0.01).Compared with the clomiphene group,the combination group showed increased FSH levels and T levels(P<0.05).Compared with the Bushen Zhongzi Decoction group,the combination group showed increased FSH levels and decreased LH/FSH,LH
