响应面法优化莲子多酚闪式提取工艺及其生物活性研究

Study on optimization of flash extraction technology of polyphenols from lotus seeds by response surface methodology and its bioactivity

摘要 以莲子为原料通过闪式提取法提取莲子多酚。以多酚提取量为指标,在单因素试验的基础上通过响应面法优化提取工艺,并研究莲子多酚的生物活性。结果表明:最佳提取工艺为液料比16∶1(mL/g)、提取时间78 s、乙醇体积分数47%,在此条件下莲子多酚提取量为(8.28±0.32)mg/g。莲子多酚对DPPH自由基和ABTS自由基的最大清除率分别为89.74%±1.15%和95.94%±1.21%,且对铁离子也有较强的还原能力;莲子多酚对α-淀粉酶和α-葡萄糖苷酶的最大抑制率分别为71.31%±1.08%和83.79%±1.11%,表明莲子多酚具有较强的抗氧化活性和降血糖活性。 Lotus seeds polyphenols were extracted by flash extraction method using lotus seeds as raw material.Using the extraction amount of polyphenols as an indicator,the extraction process was optimized by response surface method on the basis of single factor test,and the biological activity of lotus seeds polyphenols was studied.The results showed that the optimal extraction process were as follows:liquidsolid ratio 16:1(mL/g),extraction time 78 s and ethanol volume fraction 47%.Under these conditions,the extraction amount of lotus seeds polyphenols was(8.28±0.32)mg/g.The maximum scavenging rates of lotus seeds polyphenols on DPPH free radicals and ABTS free radicals were 89.74%±1.15%and 95.94%±1.21%,respectively,and it also had strong reducing ability on iron ions.The maximum inhibition rates of lotus seeds polyphenols onα-amylase andα-glucosidase were 71.31%±1.08%and 83.79%±1.11%,respectively,indicating that lotus seeds polyphenols had strong antioxidant activity and hypoglycemic activity.
