摘要
采用响应面法,以超声辅助提取的方式对头花龙胆总多酚提取工艺进行优化,并检测其多酚提取物体外降糖降脂活性。结果显示:最佳提取参数为超声时间78 min,料液比1∶81(g/mL),乙醇体积分数61%。此条件下多酚得率最高,达到2.45%,与预测得率2.42%的相对偏差为1.24%,说明此模型拟合度较好。体外生物活性检测发现,其多酚提取物对葡萄糖吸收有延迟作用,同时对胆酸盐具有结合能力。
Response surface methodology was used to optimize the extraction process of total polyphenols from Gentiana ceph-alantha Franch.ex Hemsl by ultrasonic-assisted extraction,and the in vitro hypoglycemic and lipid-lowering activity of its poly-phenol extract was tested.The results showed that the optimum extraction parameters were as follows:ultrasonic time 78min,the solid-liquid ratio 1:81(g/mL),and the ethanol volume fraction 61%.Under these conditions,the yield of polyphenols was the high-est,reaching 2.45%,and the relative deviation from the predicted yield of 2.42%was 1.24%,which indicated that the model had an excellent fitting degree.Vitro bioactivity test showed that the polyphenol extract had a delayed effect on glucose absorption and a binding ability to cholate.
