摘要
为获得可用于嗜热酯酶制备的高活性产酯酶微生物,满足果蔬制品精深加工用酶需求,本试验利用含三丁酸甘油酯的筛选培养基在堆肥高温期样品中筛选到一株高活性产嗜热酯酶菌株GRJ 2,对其进行了形态学、生理生化和分子生物学鉴定,并研究了其最适发酵条件。结果发现,经鉴定筛选到的GRJ 2菌株为嗜热淀粉芽孢杆菌(Caldibacillus thermoamylovorans)。其最适生长温度为55℃;最适生长pH值为7,pH耐受范围5~9。菌种最佳活化时间为10 h,分批培养条件下嗜热酯酶最佳收获时间为16~22 h,及时补加新鲜培养液,可将产酶期延长4 h。菌株GRJ2具有较高的产酯酶活性,酸碱耐受性强,可以用于工业酯酶的制备。
In order to obtain highly active thermophilic esterase producing microorganisms who can produce the thermophilic esterase to meet the enzyme requirements for the intensive processing of fruits and vegetables,a highly active producing thermophilic esterase strain GRJ 2 was screened from the compost in the thermophilic stage using the screening medium containing glyceryl butyrate.The strain was identified by morphology,physiology and biochemistry,and molecular biology methods.And the optimal fermentation conditions were studied.The results showed that the strain GRJ 2 was Caldibacillus thermoamylovorans.Its optimum growth temperature was 55℃and the optimal pH value was 7,whose tolerance range was wide from 5 to 9.The optimum activation time for this strain was 10 h and the optimum harvest stage of thermophilic esterase was from 16-22 h.The enzyme production period could be extended for 4 h by adding fresh culture medium in time.The strain GRJ 2 had high esterase activity and strong acid and base tolerance,which could be used for the preparation of industrial esterase.
