摘要
目的探讨飞龙掌血对大鼠动脉粥样硬化(atherosclerosis,AS)的影响及其对心血管损伤的调节和改善作用。方法采用高脂饲料联合维生素D3和L-甲硫氨酸建立AS大鼠模型。将大鼠随机分为正常组,模型对照组,飞龙掌血提取物低、中、高剂量组,阿托伐他汀组(阳性药)。于给药后第8周,对各组大鼠体质量、血脂水平、血清中的丙氨酸转氨酶(ALT)和天冬氨酸转氨酶(AST)、脂质氧化(MDA)、钙离子(Ca^(2+))、氧化低密度脂蛋白(ox-LDL)、氧化低密度脂蛋白受体1(sLOX-1)和单核细胞趋化蛋白1(MCP-1)的含量、主动脉和肝脏的病理及脂质沉积进行检测来探讨飞龙掌血对AS大鼠的影响。结果实验成功建立了大鼠动脉粥样硬化模型,飞龙掌血提取物能一定程度降低AS大鼠的体质量,降低AS大鼠血清中的血脂水平,降低AST、ALT、MDA、Ca^(2+)、ox-LDL、sLOX-1和MCP-1的含量,能改善血管动脉壁的结构紊乱、纹理不清和肝细胞脂肪变性情况,减轻大鼠主动脉和肝脏组织中脂质沉积。结论飞龙掌血提取物可通过调节血脂水平和脂质代谢紊乱、抗脂质过氧化、抑制血管钙化和调控oxLDL/LOX-1信号通路多个途径改善动脉粥样硬化大鼠的病理状况,对心脑血管疾病有一定的防治作用,为进一步探讨飞龙掌血抗动脉粥样硬化作用机制研究奠定了基础。
Objective To study the effect of Toddalia asiatica(L.)Lam on atherosclerotic rats and improvement on cardiovascular injury.Methods Establishment of atherosclerosis(AS)rat model by high fat diet combined with vitamin D,and L-methionine.The rats were randomly divided into normal group,model control group,low,medium and high dose group of Toddalia asiatica(L.)Lam extract,atorvastatin group(positive drug).At 8 weeks after administration,to explore the effect of Toddalia asiatica(L.)Lam on AS rats was investigated by detecting the weight,blood lipid level,AST,ALT,MDA,Ca^(2+),ox-LDL,sLOX-1,MCP-1,pathology and lipid deposition of aorta and liver.Results The experiment successfully established a rat atherosclerosis model.Toddalia asiatica(L.)Lam extract can improve the body weight of AS rats to a certain extent,and reduce the serum blood lipid levels,AST,ALT,MDA,Ca^(2+),ox-LDL,sLOX-1 and MCP-1 content of AS rats,and improve the structural disorder,unclear texture and hepatic steatosis of vascular artery wall,and reduce the lipid deposition in aorta and liver of AS rats.Conclusion Toddalia asiatica(L.)Lam extract can improve the pathological conditions of atherosclerotic rats by regulating blood lipid levels and lipid metabolism disorders,anti-lipid peroxidation,inhibiting vascular calcification,and regulating ox-LDL/LOX-1 signaling pathway,which has a certain preventive and therapeutic effect on cardiovascular and cerebrovascular diseases,and lays a foundation for further study on the mechanism of anti-atherosclerosis effect of Toddalia asiatica(L.)Lam extract.
