摘要
目的:建立透骨香水煎液的HPLC指纹图谱,并进行化学模式识别研究。方法:采用Kromasil^(Ⓡ)100-5 C_(18)(250 mm×4.6 mm,5μm)色谱柱;流动相为乙腈-0.15%三氟乙酸溶液,梯度洗脱;流速为0.8 mL/min;检测波长为280 nm;柱温为20℃;进样量为20μL。采用“中药色谱指纹图谱相似度评价系统(2004 A版)”计算相似度,SPSS 26.0软件进行聚类分析和主成分分析及正交偏最小二乘判别分析。结果:建立了透骨香水煎液HPLC指纹图谱,确定了19个共有峰,指认了5个成分,17批样品与对照图谱的相似度均≥0.945。聚类分析结果表明17批样品聚为3类,正交偏最小二乘判别分析发现7个成分是造成不同批次间质量差异的主要标志性成分。结论:该研究建立的HPLC指纹图谱可为透骨香质量评价提供参考。
Objective:To establish the HPLC fingerprint of water decoction of Gaultheria leucocarpa var.crenulata rhizome and conduct chemical pattern recognition research.Methods:The chromatographic separation was performed on a Kromasil^(Ⓡ)100-5 C_(18)(250 mm×4.6 mm,5μm)column,the mobile phase was acetonitrile-0.15%trifluoroacetic acid solution with gradient elution,flow rate was 0.8 mL/min,detection wavelength was 280 nm,column temperature was 20℃,injection volume was 20μL.The similarity was calculated by"Traditional Chinese medicine chromatographic fingerprint similarity evaluation system(2004 A version)",and the cluster analysis,principal component analysis and orthogonal partial least squares discriminant analysis were carried out by SPSS 26.0 statistical software.Results:The HPLC fingerprint of the water decoction of Gaultheria leucocarpa var.crenulata rhizome was established and 19 common peaks were determined,and 5 chromatographic peaks were identified.The similarity between the 17 batches of samples and the control map was≥0.945.The results of cluster analysis showed that 17 batches of samples were grouped into 3 groups.Orthogonal partial least squares discriminant analysis showed that 7 components were the main indicators of quality difference among different batches.Conclusion:The HPLC fingerprint established in this study can provide a reference for the quality evaluation of Gaultheria leucocarpa var.crenulata rhizome.
