摘要
为筛选适用于建兰(Cymbidium ensifolium)花香物质合成相关基因表达分析的内参基因,本研究从建兰‘小桃红’转录组测序结果中筛选出ACT、GAPDH、EF1α、TUA、TUB、CYP、UBQ和ADF这8个常用的内参基因作为候选内参基因。以建兰‘小桃红’的不同花期(小蕾期,中蕾期,大蕾期,初开期,盛花期,衰败期)和盛花期不同花器官(萼片,花瓣,唇瓣,合蕊柱)为实验材料,利用RT-qPCR技术分析候选内参基因在不同样品中的表达量,并结合内参基因稳定性分析软件对候选基因表达稳定性进行分析。结果表明:在候选内参基因中,TUB的稳定性最高,可作为建兰‘小桃红’花香物质合成相关基因表达分析的最适内参基因。
In order to select the appropriate reference genes for RT-qPCR analysis of the correlated genes in biosynthesis pathway of floral scent in Cymbidium ensifolium.In this study,eight commonly used reference genes,including ACT,GAPDH,EF1α,TUA,TUB,CYP,UBQ and ADF,were selected as candidate reference genes based on the C.ensifolium transcriptome data.The expression levels of candidate reference genes were detected by RT-qPCR in different flower developmental stages and different floral organs.Expression stability of candidate genes was analyzed by reference gene stability analysis programs.The results showed that TUB was characterized as the most stable gene in candidate reference genes.Therefore,TUB can be used as the most suitable reference gene for the expression analysis of the genes related to the biosynthesis pathway of floral scent in C.ensifolium.
