建兰R2R3-MYB转录因子家族鉴定及表达分析 被引量:2

Identification and Expression Analysis of the R2R3-MYB Transcription Factor in Cymbidium ensifolium

摘要 R2R3-MYB基因家族是MYB转录因子中最大的亚家族之一,参与植物根、茎、叶和花的生长发育过程。本研究基于建兰‘小桃红’花蕾和盛开花朵的转录组数据,筛选鉴定出建兰R2R3-MYB转录因子,并分析与花发育相关R2R3-MYB亚家族成员在根、叶和花开放不同时期中的表达模式。结果表明,从建兰转录组中筛选出81个MYB转录因子,其中R2R3-MYB成员有30个。30个R2R3-MYB基因主要位于细胞核内,相对分子量为12.19~58.86 kD,氨基酸序列大小为129~535 aa,等电点为4.73~9.87。亚细胞定位预测分析显示它们定位于细胞核。系统进化分析表明,建兰30个R2R3-MYB成员聚集到拟南芥12个亚家族中。将其中18个位于花发育相关亚家族下的建兰R2R3-MYB基因,在根、叶和不同花朵开放时期的样品中进行实时荧光定量分析,结果表明,不同亚家族成员在不同组织和花发育各阶段中呈现差异性表达,特别是S5、S7、S9和S18、S19亚家族成员在花发育过程中高表达。本研究详细剖析了建兰R2R3-MYB成员在花发育不同时期的表达模式,为进一步研究建兰R2R3-MYB基因在花发育过程中的潜在功能提供了理论参考。 R2R3-MYB are one of the largest subfamilies of MYB transcription factors and participate in the growth and development of plant roots,stems,leaves and flowers.In this study,R2R3-MYB transcription factor were screened and identified based on RNA-Seq data of the bud and blooming flowers in Cymbidium ensifolium'Xiaotaohong'.Furthermore,the expression patterns of C.ensifolium R2R3-MYB genes related to flower development in roots,leaves and different stages of the flower were analyzed.The results showed that 81 MYB transcription factors were selected from the transcriptome of C.ensifolium,of which thirty were R2R3-MYB members.Thirty R2R3-MYB genes were mainly located in the nucleus,with the amino acid sequence size ranging from 129 to 535 aa,a relative molecular weight of 12.19 to 58.86 kD,and protein isoelectric of 4.73 to 9.87.The phylogenetic analysis revealed that thirty C.ensifolium R2R3-MYB members could be classified into 12 subgroups.qRT-PCR was carried out to detect tissue-specific expression differences of CeMYB genes.The expression levels of S5,S7,S9,S18,and S19 subfamily members were high during the process of flower development in C.ensifolium'Xiaotaohong'.In this study,we analyzed the expression patterns of R2R3-MYB members at different stages of flower development,providing a theoretical reference for further research on the potential function of R2R3-MYB gene in flower development.
