摘要
采用紫外可见分光光度法测定火龙果中花青素的含量。通过单因素法考察显色剂混合体积比、水浴温度、缓冲溶液pH和水浴时间等样品前处理条件对花青素吸光度的影响,并经正交试验优化以上处理条件。结果表明,样品前处理的最优条件为:显色剂(香兰素甲醇溶液与盐酸甲醇溶液)混合体积比1∶0.9、水浴温度28℃、缓冲溶液pH 4.6、水浴时间23 min;测得红心火龙果的果皮、果肉与白心火龙果的果皮、果肉中花青素的含量分别为7.010、6.843 mg·(100 g)^(-1)与2.121、0.821 mg·(100 g)^(-1),相对标准偏差范围为0.03%~0.68%,精密度高。
UV visible spectrophotometry was used for the determination of the anthocyanin content in pitaya fruit.The effects of sample pretreatment conditions,such as volume ratio of vanillin methanol solution to hydrochloric acid methanol solution,water bath temperature,pH of buffer solution and water bath time were investigated by single-factor method,and the above treatment conditions were optimized by orthogonal test.The results showed that the optimal conditions for the pretreatment of the samples were:color developer(vanillin methanol solution and hydrochloric acid methanol solution)mixing volume ratio of 1∶0.9,water bath temperature of 28 ℃,buffer solution pH 4.6,water bath time of 23 min.Under this condition,the anthocyanin content in the peel and flesh of red pitaya was 7.010 and 6.843 mg·(100 g)^(-1),and the corresponding anthocyanin content in the peel and flesh of white pitaya were 2.121 and 0.821 8 mg·(100 g)^(-1),with relative standard deviation RSD ranging from 0.03% to 0.68%,and high precision.
