不同栽培模式下铁皮石斛多糖提取及抗氧化活性研究 被引量:10

Extraction optimization and antioxidant activity of polysaccharides from Dendrobium candidum under different cultivation

摘要 铁皮石斛——中国传统药材,多糖作为铁皮石斛最重要的初生代谢产物,在抗肿瘤、抗氧化、降血糖以及解酒护肝等方面具有良好的功效,在食品药品添加剂方面拥有广阔的市场前景。本研究以晋产苗床和树桩栽培的铁皮石斛为原材料,以水作为溶剂,对铁皮石斛中的多糖采取浸泡式提取法,比较两种栽培模式下的多糖提取率。详细分析提取温度、料液比、提取时间对提高多糖提取率的影响,并在此基础上采用BoxBehnken中心组合设计和响应面法建立预测模型,优选出多糖提取最佳参数条件。结果显示:苗床栽培的铁皮石斛的多糖含量略大于树桩栽培的铁皮石斛。料液比1∶35,提取时间2 h,提取温度80℃时,苗床栽培铁皮石斛多糖的提取率达65.52%±0.1%;当料液比为1∶45,提取时间为2 h,提取温度为80℃时,树桩栽培铁皮石斛多糖的提取率为52%±0.24%。两种栽培模式提得的多糖抗氧化性检测结果表明:铁皮石斛多糖清除DPPH及羟基自由基的活性均较好,苗床栽培的铁皮石斛多糖抗氧化性优于树桩栽培。 Dendrobium officinale is a traditional Chinese medicine. Polysaccharide,as the most important primary metabolite of Dendrobium officinale,has good effects on anti-tumor,anti-oxidation,hypoglycemic and antialcohol and liver protection,etc.. It has a broad market prospect in food and drug additives. In this study,Dendrobium officinale cultivated in seedling bed and tree stump were used as raw materials,and water was used as the solvent to extract polysaccharide from Dendrobium officinale. The extraction rates of polysaccharide under two cultivations were compared. The effects of three factors(extraction temperature,solid-liquid ratio and extraction time)on the extraction rate of polysaccharide were analyzed. Box-Behnken central composite design and response surface methodology were used to establish the prediction model,and optimized polysaccharide extraction process. The results showed that the polysaccharide content of Dendrobium officinale in seedling bed culture was slightly higher than that in tree stump culture.When the ratio of solid to liquid was 1∶35,the extraction time was 2 h,and the extraction temperature was 80℃,the extraction yield of polysaccharides reached 65.52%±0.16%. When the ratio of solid to liquid was 1∶45,the extraction time was 2 h,and the extraction temperature was 80℃,the polysaccharide content of Dendrobium officinale was 52%±0.24%. The antioxidant activity of polysaccharides extracted from the two cultivations showed that Dendrobium officinale polysaccharide had better scavenging activity to DPPH and hydroxyl radical,and the antioxidant activity of polysaccharides extracted from seedling bed cultivation was better than that from tree stump cultivation.
