铁皮石斛多糖对癫痫大鼠认知功能及海马神经元凋亡的影响 被引量:9

Effects of dendrobium officinale polysaccharides on cognitive function and hippocampal neuron apoptosis in epileptic rats

摘要 目的探讨铁皮石斛多糖(dendrobium officinale polysaccharides,DOP)对癫痫大鼠认知功能的作用及可能作用机制。方法收集SPF级Wistar雄性大鼠85只,随机选取15只为对照组,余大鼠给予腹腔注射毛果芸香碱建立癫痫大鼠模型,将造模成功的64只大鼠随机分为癫痫模型组、DOP低剂量组、DOP高剂量组和丙戊酸钠组,每组各16只。比较各组大鼠首次发作潜伏期的差异;采用Morris水迷宫实验检测各组大鼠逃避潜伏期、穿越平台次数;采用ELISA法检测海马组织白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)水平;采用尼氏染色和TUNEL染色观察海马组织神经元细胞存活数及凋亡情况;采用Western blot法检测海马组织核转录因子-κB(NF-κB)抑制蛋白α(IκBα)、p-IκBα、NF-κB p65、p-NF-κB p65、B细胞淋巴瘤/白血病-2基因(Bcl-2)蛋白及Bcl-2相关X蛋白(Bax)的表达。结果与癫痫模型组比较,DOP低、高剂量组及丙戊酸钠组大鼠发作潜伏期延长、逃避潜伏期缩短、穿越平台次数及神经元存活数目增加,海马组织IL-1β、TNF-α水平、神经元凋亡率降低,p-IκBα、p-NF-κB p65、Bax蛋白相对表达量降低,Bcl-2蛋白相对表达量升高(均P<0.05);DOP高剂量组与丙戊酸钠组间大鼠发作潜伏期、逃避潜伏期、穿越平台次数、神经元存活数目、海马IL-1β和TNF-α水平、神经元凋亡率以及p-IκBα、p-NF-κB p65、Bax、Bcl-2蛋白相对表达水平比较均无统计学差异(均P>0.05)。结论DOP可改善癫痫大鼠学习认知能力,减轻炎症反应,降低海马神经元细胞凋亡率,其作用机制可能与抑制NF-κB通路有关。 Objective To explore the improvement effect of dendrobium officinale polysaccharide(DOP)on the cognitive function of epileptic rats and its possible mechanism.Methods A total of 85 SPF Wistar male rats were collected,and 15 rats were randomly selected as the control group.The remaining rats were intraperitoneally injected with pilocarpine to establish an epilepsy rat model.The 64 successfully modeled rats were randomly divided into an epilepsy model group,a DOP low dose group,a DOP high dose group,and a sodium valproate group,with 16 rats in each group.We compared the difference in the latency of the first attack recorded in each group of rats.The Morris water maze test was used to detect the latency period and the number of platform crossings,the levels of interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α)in hippocampus were detected by ELISA.Nissal and TUNEL staining were used to observe the number of neuronal cells and apoptosis of neuronal cells.Western blot was used to detect the hippocampal NF-κB inhibitor protein alpha(IκBα)and phosphorylation(p-)IκBα,NF-κB p65,p-NF-κB p65,B-cell lymphoma/leukemia-2 gene(Bcl-2),Bcl-2 assoaciated X protein(Bax)protein expression.Results Compared with the epilepsy group,the onset latency was prolonged,the escape latency was shortened,the number of crossing platforms and the number of neuron survival increased,the levels of IL-1βand TNF-αin the hippocampus,and the apoptosis rate of neurons decreased,the relative expression of p-IκBα,p-NF-κB p65,Bax protein decreased,and the relative expression of Bcl-2 protein increased in DOP low dose group,DOP high dose group,and sodium valproate group(all P<0.05).There were no significant difference in the onset latency,the escape latency,the number of crossing platforms and the number of neuron survival,the levels of IL-1βand TNF-αin the hippocampus,the apoptosis rate of neurons,the relative expression of p-IκBα,p-NF-κB p65,Bax and Bcl-2 protein between the DOP high dose group and the sodium valproate group(all P
