摘要
目的研究结直肠癌组织中同源盒A10(HOXA10)和P53蛋白表达与临床病理特征及预后的关系。方法收集2017年4月至2019年10月在西安市中心医院行手术治疗的97例结直肠癌患者的组织标本,采用免疫组织化学染色法检测癌组织及对应癌旁正常组织中HOXA10和P53蛋白表达情况。分析HOXA10和P53蛋白表达与临床病理特征的关系;Spearman相关性分析结直肠癌组织中HOXA10和P53表达的相关性;Kaplan-Meier生存曲线分析HOXA10和P53蛋白表达与患者术后生存率的关系。结果97例结直肠癌组织中HOXA10和P53蛋白阳性表达率为74.2%和68.0%,对应癌旁正常组织中阳性表达率为35.1%和29.9%,差异均有统计学意义(P<0.001)。结直肠癌组织中HOXA10和P53蛋白表达呈正相关(r=0.657,P<0.05)。结直肠癌组织中HOXA10蛋白表达与TNM分期、分化程度、淋巴结转移及浸润深度有关,P53蛋白表达与肿瘤直径、TNM分期、分化程度、淋巴结转移、脉管浸润及浸润深度有关,差异均有统计学意义(P<0.05)。随访32(5~61)个月,HOXA10和P53蛋白阳性表达患者术后5年累积生存率为45.8%和42.4%,显著低于HOXA10和P53蛋白阴性表达患者的68.0%和64.5%,差异有统计学意义(χ^(2)=3.425,P=0.039;χ^(2)=3.087,P=0.043)。结论HOXA10和P53蛋白阳性表达患者临床分期更高、分化程度更低,伴淋巴结转移的风险增加,与结直肠癌的发生发展、浸润转移和预后有关。
Objective To investigate the relationship between homeobox A10(HOXA10)and P53protein expression and clinicopathological features and prognosis in colorectal cancer(CRC).Methods The clinical tissue samples of 97 patients with CRC who underwent surgical treatment in Xi’an Central Hospital from April 2017 to October 2019 were collected.Immunohistochemical staining was used to detect the protein expression of HOXA10 and P53 in cancer tissues and corresponding adjacent normal tissues.The relationship between HOXA10 and P53 expression and clinicopathological features was analyzed.Spearman correlation analysis was used to analyze the relationship between HOXA10 and P53 expression in cancer tissues.Kaplan-Meier survival curves were used to analyze the relationship between HOXA10 and P53protein expression and postoperative survival rate.Results The positive expression rates of HOXA10 and P53 in 97 cancer tissues were 74.2%and 68.0%,corresponding to the positive expression rates of 35.1%and 29.9%in adjacent normal tissues,and the difference were statistically significant(P<0.001).There was a positive correlation between HOXA10 and P53 protein expression in CRC(r=0.657,P<0.05).The expression of HOXA10 protein in cancer tissues was correlated with TNM stage,differentiation degree,lymph node metastasis and depth of invasion,and the expression of P53 protein was closely related to tumor diameter,TNM stage,differentiation degree,lymph node metastasis,vascular invasion and invasion depth(all P<0.05).After a follow-up of 32(5-61)months,the 5-year cumulative survival rates of patients with positive HOXA10 and P53 protein expression were 45.8%and 42.4%,significantly lower than 68.0%and 64.5%of patients with negative HOXA10 and P53 protein expression(χ^(2)=3.425,P=0.039;χ^(2)=3.087,P=0.043).Conclusion Patients with positive expression of HOXA10 and P53 proteins have higher clinical stage,lower differentiation degree and increased risk of lymph node metastasis,which is related to the occurrence,development,invasion,metastasis and
