摘要
为明确甘肃省静宁县苹果果实褐腐病病原菌种类,对采自该地区苹果果实褐腐病病原菌种类进行鉴定。通过组织分离法将菌株分离纯化后,采用柯赫氏法则进行致病性验证,结合形态学特征和内转录间隔区基因(Internal Transcribed Spacer,ITS)、β-微管蛋白基因(β-Tubulin,TUB2)和Laccase(Lcc2)多基因的系统发育分析以及常规PCR测定对分离菌株进行鉴定。结果表明,分离菌株为云南链核盘菌(Monilia yunnanensis),可同时侵染不同品种果实,致病性结果显示对‘秦冠’‘富士’‘新红星’有致病性,而对‘嘎啦’致病性较弱。
Brown rot is one of the most economically important diseases of Malus spp.Mill.In order to identify the pathogen of brown rot of apples in Jingning county of Gansu province.In this study,samples of brown rot disease were collected and purified by tissue isolation.The Koch’s rule was used for pathogenicity tests.Combined with morphological characteristics,and phylogenetic analysis based on internal transcribed spacer,β-tubulin and laccase(Lcc2),and conventional PCR were used to identify isolates.The results showed that the isolated strain was Monilia yunnanensis,and the pathogenicity results showed that it was pathogenic to‘Qinguan’‘Fuji’and‘Starkrimson’,but not to‘Gala’.Monilia yunnanensis was identified as the local apple brown rot pathogen.This study provides a theoretical basis for further control of apple brown rot.
