Determination of 18 Amino Acids in Edible Bird’s Nest Using UPLC-MS/MS

摘要 应用超高效液相色谱-串联质谱联用技术测定燕窝中的氨基酸含量。应用盐酸水解和碱性水解法处理燕窝样品,超高效液相-质谱/质谱联用仪(UPLC-MS/MS)测定燕窝中氨基酸含量及其组成成分,以不同取样量考察方法准确度与精密度。结果表明:该方法的相对偏差在0.01%~13.6%之间, SRSD在0.85%~4.98%之间, 30批干燕窝中18种氨基酸总量在59.10~68.73 g/100 g之间;必需氨基酸含量20.91~25.57 g/100 g,必需氨基酸占总氨基酸比例35.3%~39.60%,必需氨基酸总量占氨基酸总量约37.0%。该方法简单快速,灵敏度高,可为高蛋白质食品中氨基酸的快速高效检测提供参考依据。 To analyze the amino acids in edible bird’s nest using UPLC-MS/MS,the edible bird’s nest was treated by hydrochloric acid hydrolysis and alkaline hydrolysis,and the amino acid contents and components in bird’s nest were determined by UPLC-MS/MS,examination of method accuracy and precision with different sample sizes.The relative deviation of this method was 0.01%-13.6%;The Swas between 0.85% and 4.98%;The total amount of 18 amino acids in 30 batches of dried bird’s nest were 59.10-68.73 g/100 g;The essential amino acid content was 20.91-25.57 g/100 g;Essential amino acids accounted for 35.3% to 39.60% of the total amino acids;The total essential amino acids accounted for about 37.0% of the total amino acids.The method was simple,rapid and sensitive,which could provide a reference for the rapid and efficient determination of amino acids in high protein foods.
