摘要
采用毛细区带管电泳测定竹荪加工过程中产生的菌盖、菌托废弃物中的人体必需氨基酸组成和含量,经2-(11H-苯[a]咔唑)乙基氯甲酸酯柱前衍生的8种人体必需氨基酸在11 min内可实现快速高效基线分离。该方法检出限(按信噪比S/N=3)为0.77~1.56 mmol/L,线性范围为5~100 mmol/L,各氨基酸峰面积相对标准偏差均小于2.2%,迁移时间相对标准偏差均小于1.6%,方法的重现性较好;测定竹荪废弃物中氨基酸的回收率93.5%~101.2%,表明该方法对于分析物的同步测定是比较准确的。通过建立的方法具有线性范围宽、重现性好、快速高效等特点,可用作竹荪废弃物中氨基酸组成测定的常规方法。
Capillary zone tube electrophoresis was used to determine the composition and content of human essential amino acids in the caps and cedar waste produced during the processing of bamboo fungus.After 2-(11 H-benzene[a]carbazole)ethyl chloroformate column The 8 essential amino acids derived from the previous can achieve rapid and efficient baseline separation within 11 minutes.The detection limit of the method(according to the signal-to-noise ratio S/N=3)is 0.77~1.56 mmol/L,the linear range is 5~100 mmol/L,the relative standard deviation of each amino acid peak area is less than 2.2%,and the migration time is relatively The standard deviations are all less than 1.6%,and the method has good reproducibility.The recovery rate of amino acid in the waste of bamboo fungus is 93.5%~101.2%,which shows that the method is relatively accurate for the simultaneous determination of analytes.The established method has the characteristics of wide linear range,good reproducibility,rapid and high efficiency,etc.,and can be used as a routine method for determination of amino acid composition in dictyophora wastes.
