摘要
目的探讨类风湿关节炎(rheumatoid arthritis,RA)专利中药复方的用药规律及相关机制,为中医药治疗RA及新药研发提供参考。方法检索国家知识产权局中国专利公布网站中治疗RA的专利中药复方数据,运用IBM SPSS Statistics 25.0软件进行药物频数统计及系统聚类分析。运用IBM SPSS Modeler 18.0软件中Apriori关联规则进行建模分析。通过Cytoscape 3.6.1构建中药共现核心复杂网络,筛选出联接紧密的中药集群,并借助该软件中的ClueGO插件对新药物组合进行KEGG通路富集分析。结果共纳入中药复方686个,涉及药物1035味,高频药物有当归(217次,31.68%)、防风(148次,21.61%)、独活(145次,21.17%)、桂枝(141次,20.58%)等;药物类别以祛风湿药、活血化瘀药、补虚药为主;常用药对有羌活-独活、当归-黄芪、草乌-川乌等;药组有防风-羌活-独活、当归-羌活-独活;网络聚类得到4个新的药物组合:①独活、羌活、防风、当归、川芎、红花、牛膝、杜仲、木瓜、秦艽、续断;②桂枝、白芍、黄芪、白术、麻黄、甘草、鸡血藤、伸筋草、威灵仙、苍术、防己;③青风藤、生地黄、赤芍、丹参;④全蝎、穿山甲、地龙、乌梢蛇、川乌;新的药物组合通路分析得到丝裂原活化蛋白激酶(MAPK)信号通路、白细胞介素17(IL-17)信号通路、缺氧诱导因子1(HIF-1)信号通路等8条共同信号通路。结论国家专利中药复方治疗RA的主要治法为祛风除湿、活血化瘀和补益肝脾肾,并善用虫类药剔络搜邪。新的药物组合可能通过多成分、多靶点调控MAPK、IL-17、HIF-1等信号通路发挥治疗作用。
Objective To explore the medication rules and mechanism of patented traditional Chinese medicine(TCM)compounds for rheumatoid arthritis(RA),and to provide reference for the research and development of TCM new drugs for clinical treatment of rheumatoid arthritis.Methods All patented compound TCM prescriptions related to the treatment of RA were searched from the national patent database.IBM SPSS Statistics 25.0 software was used for drug frequency statistics and systematic cluster analysis.Apriori association rules in IBM SPSS Modeler 18.0 software were used for modeling analysis.The co-occurrence core complex network of TCMs was constructed by Cytoscape 3.6.1,to select the closely linked TCM cluster;and KEGG pathway enrichment analysis of the new TCM combinations was performed with the help of the ClueGO plugin in the software.Results A total of 686 Chinese herbal compounds were included,involving 1035 herbs.The high-frequency herbs were Angelicae sinensis radix(217 times,31.68%),Saposhnikoviae radix(148 times,21.61%),Angelicae pubescentis radix(145 times,21.17%),Cinnamomi ramulus(141 times,20.58%),etc.The drug categories of expelling wind and dampness drugs,promoting blood circulation and removing blood stasis drugs,tonifying deficiency drugs were predominant.The commonly used drug pairs were Notopterygii rhizoma et radix-Angelicae pubescentis radix,Angelicae sinensis radix-Astragali radix,Aconiti kusnezoffii radix-Aconiti radix,etc.Drug groups are Saposhnikoviae radix-Notopterygii rhizoma et radix-Angelicae pubescentis radix,Angelicae sinensis radix-Notopterygii rhizoma et radix-Angelicae pubescentis radix.Network clustering resulted in four new drug combinations:①Angelicae pubescentis radix,Notopterygii rhizoma et radix,Saposhnikoviae radix,Angelicae sinensis radix,Chuanxiong rhizoma,Carthami flos,Achyranthis bidentatae radix,Eucommiae cortex,Chaenomelis fructus,Gentianae macrophyllae radix,and Dipsaci radix.②Cinnamomi ramulus,Paeoniae radix alba,Astragali radix,Atractylodis macrocephalae rhizoma,Ephedrae
