摘要
目的 探讨系统性红斑狼疮(Systemic Lupus Erythematosus,SLE)专利中药复方的用药规律,为中医药治疗SLE及新药研发提供参考。方法 检索国家知识产权局中国专利公布网站中治疗SLE的专利中药复方数据,运用Microsoft Office Excel 2019与IBM SPSS Modeler 18.0软件进行药物频数统计,以Apriori关联规则进行建模分析,Cytoscape 3.6.1软件构建中药核心复杂网络,IBM SPSS Statistics 25对核心网络中的药物进行系统聚类分析,并利用Cytoscape 3.6.1软件的ClueGO插件对新的中药组合进行KEGG通路富集分析。结果 共纳入中药复方194个,涉及药物537味,高频中药有生地黄(79次,40.72%)、黄芪(58次,29.90%)、甘草(53次,27.32%)、牡丹皮(50次,25.77%)等;药物类别以清热药、补虚药和活血化瘀药为主;药物性味以寒、苦居多,归经多为肝、肺、肾、脾经;常用药对有生地黄-牡丹皮、生地黄-当归、生地黄-赤芍等;药组有牡丹皮-赤芍-生地黄、生地黄-赤芍-牡丹皮;网络聚类得到4个新的中药组合:牡丹皮、水牛角、生地黄、赤芍、紫草、白花蛇舌草;牛膝、黄芩、茯苓、泽泻、甘草;黄连、连翘、金银花;丹参、鸡血藤、党参、当归、女贞子、黄芪、白术、枸杞子、墨旱莲;新的中药组合通路分析得到缺氧诱导因子1(HIF-1)信号通路、肿瘤坏死因子(TNF)信号通路、Toll样受体信号通路等15条信号通路。结论 国家专利中药复方治疗SLE的主要治法为清热凉血解毒、健脾利湿和补益肝肾,并以活血化瘀法贯穿其中。新的中药组合可通过多成分、多靶点调控HIF-1、TNF、Toll样受体等信号通路发挥治疗作用。
Objective To explore the medication law of traditional patented Chinese medicine compound for treating systemic lupus erythematosus, and to provide reference for the research and development of TCM new drugs for clinical treatment of systemic lupus erythematosus. Methods All compound TCM patents related to the treatment of systemic lupus erythematosus were searched from the national patent database(http://epub.sipo.gov.cn). Microsoft Office Excel 2019 and IBM SPSS Modeler 18.0 software were used for drug frequency statistics,Apriori association rules were used for modeling analysis,Cytoscape 3.6.1 was used to construct the core complex network of traditional Chinese medicine,IBM SPSS Statistics 25 was used for systematic cluster analysis of drugs in the core network,and ClueGO plugin of Cytoscape 3.6.1 software was used for KEGG pathway enrichment analysis for new TCM combinations. Results A total of 194 compound TCM patents were included,involving 537 Chinese medicine. Highfrequency herbs included Rehmanniae radix(79 times,40.72%),Astragali radix(58 times,29.90%),Glycyrrhizae radix et rhizoma(53 times,27.32%),Moutan cortex(50 times,25.77%),etc. The top three frequently appeared drug categories were heat-clearing medicine, deficiency-tonifying medicine and blood-circulation activating and blood-stasis dissipating medicine. The top drug property and taste were cold and bitterness. The top meridian tropisms were liver, lung, kidney and spleen meridians. The most commonly used herbal pairs were Rehmanniae radix-Moutan cortex,Rehmanniae radix-Angelicae sinensis radix,and Rehmanniae radix-Paeoniae radix rubra,etc. Herbal combinations were Moutan cortex-Paeoniae radix rubra-Rehmanniae radix,Rehmanniae radix-Paeoniae radix rubra-Moutan cortex,etc. Network clustering resulted in four new drug combinations:‘Moutan cortex,Bubali cornu, Rehmanniae radix, Paeoniae radix rubra, Arnebiae radix, Hedyotidis diffusae herba’,‘Achyranthis bidentatae radix,Scutellariae radix,Poria,Alismatis rhizoma,Glycyrrhizae radix et rhizom
