酵母相和菌丝相克柔念珠菌诱导大鼠乳腺炎症的病理特征研究 被引量:1

Inflammatory pathological characteristics of rat mammary gland induced by yeast phase and hyphae phase of Candida krusei

摘要 为了明晰酵母相和菌丝相克柔念珠菌分别感染乳腺引起的病理特征,建立了克柔念珠菌双相感染大鼠乳腺炎症模型,观察乳腺组织的病理变化特征并分析炎症因子在乳腺中的表达水平,为病理诊断真菌性乳腺炎提供依据。研究发现:与空白对照组相比,模型组乳腺组织腺泡扩张,腺泡上皮细胞增生,乳汁分泌物消失,各种炎性细胞浸润,小叶间结缔组织减少或消失;乳腺组织匀浆中的IL-1β、TNF-α、IL-6、IL-8、IL-18质量分数呈现不同程度的升高;血清白蛋白质量浓度呈现不同程度的下调,转铁蛋白质量浓度呈现不同程度的升高。JM-1组(1×10^(7)CFU/m L)、JM-2组(1×10^(8)CFU/m L)、JM-3组(1×10^(9)CFU/m L)引发乳腺组织的损伤程度基本一致,说明酵母相克柔念珠菌各浓度都适合,均宜造模。JS-1组(1×10^(7)CFU/m L)、JS-2组(1×10^(8)CFU/m L)、JS-3组(1×10^(9)CFU/m L)均能引发乳腺组织的损伤,但JS-3组乳腺组织的病变很严重,大部分乳腺腺泡和小叶间的结缔组织消失不见,形成肉芽肿。结果表明:成功建立了克柔念珠菌双相大鼠乳腺炎模型,用酵母相建立大鼠乳腺炎模型的适宜浓度为1×10^(9)CFU/m L,用菌丝相建立大鼠乳腺炎模型的适宜浓度为1×10^(8)CFU/m L。酵母相和菌丝相克柔念珠菌感染均可以导致大鼠乳腺发生慢性炎性反应,细胞因子TNF-α、IL-1β、IL-6、IL-8、IL-18和血清白蛋白、转铁蛋白共同参与了克柔念珠菌双相感染大鼠乳腺引发的炎性损伤,其中菌丝相引发的炎性损伤迟缓但损伤程度强于酵母相。 In order to clarify the pathological features of the mammary gland caused by the yeast phase and hyphae phase of Candida krusei,a rat mammary inflammation model was established with C.krusei biphase infection,the pathological changes of the mammary tissue were observed and the expression level of inflammatory factors in the mammary gland was analyzed,providing a basis for the pathological diagnosis of fungal mastitis.The results showed that,compared with blank control group,the acinar tissue of mammary gland tissue in model group expanded,acinar epithelial cells prolifulated,milk secretions was disappeared,various inflammatory cells infiltrated,interlobular connective tissue decreased or disappeared.The mass fraction of IL-1β,TNF-α,IL-6,IL-8 and IL-18 in breast tissue homogenate were increased to different degrees.Serum albumin mass concentration showed different degrees of downregulation,transferrin mass concentration showed different degrees of increase.JM-1 group(1×10^(7)CFU/m L),JM-2 group(1×10^(8)CFU/m L),and JM-3 group(1×10^(9)CFU/m L)induced the same degree of mammary tissue damage,indicating that all concentrations of C.krusei yeast phase were suitable for modeling.JS-1 group(1×10^(7)CFU/m L),JS-2 group(1×10^(8)CFU/m L)and JS-3 group(1×10^(9)CFU/m L)can all cause the injury of breast tissue,but the pathological changes of breast tissue in JS-3 group were very serious,and most of the connective tissue between the acinus and lobules of the breast disappeared,forming granuloma.The results showed that the model of mastitis in rats was successfully established.The suitable concentration of mastitis model in rats was 1×10^(9)CFU/m L for yeast phase and 1×10^(8)CFU/m L for hyphae phase.Yeast phase and hyphae phase infection of C.krusei can lead to chronic inflammatory response in the mammary gland of rats.Cytokines TNF-α,IL-1β,IL-6,IL-8,IL-18,serum albumin and transferrins are involved in the inflammatory damage caused by C.krusei biphasic infection in the mammary gland of rats.Among them,hyphae phas
