miR-802、TNF-α在妊娠期糖尿病患者血清和胎盘组织中的表达及意义 被引量:2

The expression and clinical significance of miR-802 and tumor necrosis factor-α in serum and placenta tissue of patients with gestational diabetes

摘要 目的探究微小RNA-802(miR-802)、肿瘤坏死因子-α(TNF-α)在妊娠期糖尿病(GDM)患者血清、胎盘组织中的表达水平及临床意义。方法选取经口服葡萄糖耐量试验(OGTT)(24~28周)诊断为GDM孕妇82例为研究组(GDM组),并选取经OGTT判定为正常糖耐量(NGT)孕妇82例为对照组(NGT组)。比较NGT组与GDM组一般资料及血清、胎盘组织中miR-802、TNF-αmRNA表达水平;分析GDM孕妇血清、胎盘组织miR-802、TNF-αmRNA表达水平与空腹胰岛素(FINS)、三酰甘油(TG)、空腹血糖(FBG)、稳态模型胰岛素抵抗指数(HOMA-IR)的相关性,及miR-802表达水平与TNF-αmRNA的相关性。结果GDM组FINS、TG、FBG、HOMA-IR水平高于NGT组(P<0.05);GDM组血清、胎盘组织中miR-802、TNF-αmRNA表达水平均高于NGT组(P<0.05);GDM孕妇血清、胎盘组织miR-802、TNF-αmRNA表达水平与HOMA-IR均呈正相关(P<0.05),血清、胎盘组织miR-802表达水平与TNF-αmRNA均呈正相关(P<0.05)。结论GDM患者血清、胎盘组织miR-802、TNF-αmRNA表达水平较高,均与胰岛素抵抗(IR)显著有关,miR-802可能是治疗GDM的潜在靶点。 Objective To investigate the expression levels and clinical significance of microRNA-802(miR-802) and tumor necrosis factor-α(TNF-α) in serum and placenta tissues of patients with gestational diabetes(GDM). Methods Eighty-two pregnant women with GDM determined by oral glucose tolerance test(OGTT)(24-28 weeks) were selected for the study(GDM group), and 82 pregnant women with normal glucose tolerance(NGT) determined by OGTT were selected for control study(NGT group). The general information, miR-802 and TNF-α mRNA in serum and placenta tissue were compared between the NGT group and the GDM group. The correlations between the expression levels of miR-802 and TNF-α mRNA in serum, placental tissues of GDM pregnant women and fasting insulin(FINS), triacylglycerol(TG), fasting blood glucose(FBG), homeostasis model insulin resistance index(HOMA-IR), and the correlation between expression levels of miR-802 and TNF-α mRNA were analyzed. Results The levels of FINS, TG, FBG and HOMA-IR in the GDM group were significantly higher than those in the NGT group(P<0.05). The expression levels of miR-802 and TNF-α mRNA in serum and placental tissues of GDM group were significantly higher than those of NGT group(P<0.05). The expression levels of miR-802 and TNF-α mRNA in serum and placental tissues of GDM pregnant women were positively correlated with HOMA-IR(P<0.05), and the expression level of miR-802 in serum and placental tissues was positively correlated with TNF-α mRNA(P<0.05). Conclusion The high levels of miR-802 and TNF-α mRNA expression in serum and placental tissues of GDM patients are significantly correlated to insulin resistance(IR). And miR-802 may be a potential target for the treatment of GDM.
