摘要
目的探讨姜黄素通过Notch1途径影响前列腺癌PC-3细胞体外生长的分子机制。方法培养前列腺癌PC-3细胞,分别给予不同浓度的姜黄素处理,分别为500 nmol/L组、10μmol/L组、100μmol/L组和空白对照组。观察PC-3细胞增殖和凋亡的变化,并分析姜黄素对Notch1和PI3K mRNA水平的影响。结果不同剂量的姜黄素均可抑制PC-3细胞的增殖,诱导细胞凋亡,增加Caspase-3和Caspase-9活性(均P<0.05),并具有一定的剂量依赖性。PC-3细胞中Notch1和PI3K高表达,经过姜黄素处理的PC-3细胞,可以显著降低Notch1和PI3K mRNA水平和蛋白表达(P<0.05),体现一定的剂量依赖性。结论姜黄素通过Notch1途径抑制前列腺癌PC-3细胞的体外生长。
Objective To explore the effect of curcumin on PC-3 cell growth in vitro.Methods PC-3 cells were treated with different concentration of curcumin to observe the changes on cell proliferation,cell apoptosis and detect the effect of curcumin on Notch1 and PI3K mRNA level in PC-3 cell.Results Cell proliferation were reduced by curcumin which could also enhance the cell apoptosis by increasing Caspase-3 and Caspase-9 activity with the dose dependent manner(P<0.05,n=6).Higher Notch1,PI3K mRNA level and protein expression were obtained in PC-3 cell which could be reduced by curcumin application with the dose dependent manner(P<0.05,n=6).Conclusion Effect of curcumin regulate PC-3 cell growth negatively in vitro.
