摘要
目的:观察电针对高脂饮食诱导肥胖(DIO)大鼠脂肪组织中mTOR/P70S6K信号通路及自噬相关因子表达的影响,探讨电针减重的外周作用机制。方法:将60只雄性Wistar大鼠中随机分为正常组(10只)和高脂组(50只),采用高脂饮食复制DIO模型。将造模成功的30只大鼠随机分为模型组、电针组和假电针组,每组10只。电针组干预中脘、天枢、足三里和三阴交穴,频率2 Hz/15 Hz,强度0.5~1 mA,留针30 min,假电针组取所选穴位向外旁开5 mm处浅刺,每周连续干预5次,共治疗4周。记录各组大鼠干预前后体质量,干预后白色脂肪组织(WAT)含量、脂肪细胞面积和直径,免疫组化法检测WAT中mTOR、P70S6K的表达,RT-PCR、Western blot法分别检测mTOR、P70S6K、Beclin-1、p62的mRNA及蛋白表达。结果:与正常组比较,模型组大鼠体质量、WAT重量显著增加(P<0.01),脂肪细胞直径和面积显著增大(P<0.05),WAT中Beclin-1 mRNA及蛋白表达显著增加(P<0.01),p62、mTOR、P70S6K mRNA及蛋白表达均显著降低(P<0.01);与模型组、假电针组比较,电针组大鼠体质量、WAT重量显著减轻(P<0.01,P<0.05),脂肪细胞直径和面积显著减小(P<0.01,P<0.05),WAT中Beclin-1 mRNA及蛋白表达显著降低(P<0.01,P<0.05),p62、mTOR、P70S6K mRNA及蛋白表达均显著增加(P<0.01,P<0.05)。结论:电针可以激活WAT中mTOR/P70S6K信号通路,抑制脂肪组织过度自噬,这可能是电针实现减重效应的部分外周机制。
Objective:To observe the effects of electroacupuncture on the expression of mTOR/P70S6K signal pathway and autophagy-related factors in adipose tissue of high-fat diet induced obesity(DIO)rats,and to explore the peripheral mechanism of electroacupuncture weight loss.Methods:Sixty male Wistar rats were randomly divided into normal group(10 rats)and high-fat group(50 rats).Thirty rats were randomly divided into model group,electroacupuncture(EA)group and sham electroacupuncture group,with 10 rats in each group.The electroacupuncture group alternately intervened at bilateral Zhongwan(CV 12),Tianshu(ST 25),Zusanli(ST 36)and Sanyinjiao(SP 6)points,with a frequency of 2 Hz/15 Hz,an intensity of 0.5 to 1 mA,and kept the needle for 30 min.The sham electroacupuncture group took the selected points and opened a shallow thorn at 5 mm outward.The body mass,white adipose tissue(WAT)content,adipocyte area and diameter were measured before and after the intervention.The expressions of mTOR and P70S6K in WAT were detected by immunohistochemistry.The mRNA and protein expressions of mTOR,P70S6K,Beclin-1 and p62 in WAT were detected by RT-PCR and Western blot.Results:Compared with the normal group,the body weight and WAT weight of the model group increased(P<0.01),the diameter and area of adipocytes increased(P<0.05),the expression of Beclin-1 mRNA and protein in wat increased(P<0.01),and the expression of p62,mTOR,P70S6K mRNA and protein decreased(P<0.01).Compared with model group and sham EA group,the body weight and WAT mass of rats in EA group decreased(P<0.01,P<0.05),the diameter and area of adipocytes decreased(P<0.01,P<0.05),the expression of Beclin-1 mRNA and protein in wat decreased(P<0.01,P<0.05),and the expression of p62,mTOR,P70S6K mRNA and protein increased(P<0.01,P<0.05).Conclusion:EA could activate mTOR/P70S6K signaling pathway in WAT and inhibit excessive autophagy of adipose tissue,which may be part of the peripheral mechanism of EA to exert weight loss effect.
