黄芩对营养性肥胖大鼠血糖血脂的作用及其机理研究 被引量:2

摘要 目的:探讨黄芩对营养性肥胖大鼠血糖血脂的作用及其机理。方法:采用高脂饲料诱导SD大鼠成肥胖型大鼠,然后灌胃黄芩水提液进行干预,检测体重、体长、Lee’s指数和血糖血脂,并进行肝脏组织转录表达谱测序,评价黄芩影响肥胖大鼠血糖血脂水平变化差异基因和相关通路,并探讨其可能的作用机制。结果:黄芩可降低血糖(P<0.05),升高高密度脂蛋白胆固醇(P<0.05),而总胆固醇和低密度脂蛋白胆固醇水平均有下降趋势(P>0.05)。肝组织表达谱结果显示,相对于模型组,黄芩干预后,上调基因23个,下调基因15个(P<0.05),这些差异基因主要富集于生物节律和支链氨基酸合成代谢通路。结论:黄芩具有降低营养性肥胖大鼠血糖和血脂的作用,其可能的机制涉及影响生物节律和支链氨基酸合成代谢通路相关的基因。 Objective:To investigate the effect of serum glucose and lipid profiles in obese rats intervened by scutellariae radix.Methods:SD rats were induced into nutritional obese rats by high fat diet.Then,we gave the water extracts from scutellariae radix to the model and tested the body weight,body length,Lee’s index and serum glucose and lipid profiles.Finally,to evaluate the mechanism on changing of serum glucose and lipid profiles associated differential genes and pass way in model group intervened by scutellariae radix,we carried out liver tissue transcriptional expression experiment.Result:The serum glucose level lowered,high density lipoprotein cholesterol elevated,(P<0.05),while the total cholesterol and low density lipoprotein cholesterolshowed decreasing trend(P>0.05)after five weeks during the scutellariae radix intervened the obesity rat model.Our data from liver tissue transcriptional expression showed that 23 genes were upregulated and 15 genes were down-regulated intervened by the scutellariae radix compared with the model group.These differentially expressed genes were mainly enriched in the circadian rhythm and branched amino acid synthesis metabolic pathways(P<0.05).Conclusion:The scutellariae radixhad the effect of reducing the level of serum glucose and lipid profiles in the diet-induced obese rats and its possible mechanism involving the genes that affect the circadian rhythm and the amino acid synthesis metabolic pathways.
