电针对胰岛素抵抗大鼠认知功能与下丘脑PI3K、Akt2蛋白表达的影响 被引量:5

Effect of Electro-acupuncture on Cognitive Function and Expression Levels of Hypothalamic PI3K and Akt2 in Insulin Resistance Model Rats

摘要 目的:观察电针对胰岛素抵抗模型大鼠学习记忆能力与下丘脑中PI3K、Akt2及其磷酸化蛋白表达的影响。方法:将60只Wistar大鼠随机分为正常组、模型组、预防组和电针组,以标本配穴法电针治疗。检测各组大鼠FBG、FIN;空间学习记忆能力;下丘脑Aβ1-42阳性表达、PI3K、Akt2及其磷酸化蛋白表达。结果:(1)较正常组大鼠,模型组大鼠FBG、FINS、ISI升高,IRI降低(P<0.01);定位航行与空间探索时间延迟,跨越次数减少(P<0.01);下丘脑Aβ1-42阳性表达减少,PI3K、Akt2及其磷酸化蛋白表达减少(P<0.01)。(2)较模型组大鼠,预防组和电针组大鼠FBG、FIN、IRI降低,ISI升高(P<0.01);定位航行与空间探索时间缩短,跨越平台次数增加(P<0.01);下丘脑Aβ1-42阳性表达增加,PI3K、Akt2及其磷酸化蛋白表达增加(P<0.01)。(3)较电针组大鼠,预防组大鼠FBG、FIN、IRI降低,ISI升高(P<0.01);定位航行时间缩短(P<0.01);下丘脑PI3K、Akt2及其磷酸化蛋白表达增加(P<0.01)。结论:电针改善胰岛素抵抗模型大鼠学习记忆能力的作用机制可能与改善中枢胰岛素抵抗状态相关。 Objective:To observe the electro-acupuncture effect on cognitive function and the hypothalamic PI3 K,Akt and the phosphorylated protein expression levels of insulin resistance model rats.Methods:Sixty Wistar rats were divided into normal group,model group,protecting electro-acupuncture group and electro-acupuncture group randomly,15 rats in each group.Blood glucose levels and FIN were detected before and after treatment.Aβ1-42 expression was detected by ELISA method and protein levels of hypothalamic PI3 K and Akt were detected via western-blotting method.Results:Compared with normal group,FBG,FINS and ISI raised in model group and IRI reduced(P<0.01).The time in spatial navigation and space exploration delayed(P<0.01).Aβ1-42 expression,protein levels of hypothalamic PI3 K,Akt and the phosphorylated protein expression decreased(P<0.01).Compared with model group,FBG,FINS and ISI reduced in P-EA group and EA group and IRI increased(P<0.01) and the time in spatial navigation and space exploration was shortened(P<0.01).Aβ1-42 expression,protein levels of hypothalamic PI3 K,Akt and the phosphorylated protein expression increased(P<0.01).Compared with EA group,FBG,FINS and ISI reduced in P-EA group and IRI increased(P<0.01)and the time in spatial navigation was shortened(P<0.01).The protein levels of hypothalamic PI3 K,Akt and the phosphorylated protein expression decreased(P<0.01).Conclusion:The mechanism of electro-acupuncture improvement on learning and memory ability in insulin resistance model rats,is pertaining to the regulation of electro-acupuncture on hypothalamus insulin sensitivity in model rats.
