摘要
为进一步明确南疆地区鹅喉羚(Gazella subgutturosa)的遗传分化、遗传多样性水平和系统进化地位,以鹅喉羚南疆亚种(G.s.yarkandensis)为研究对象,运用PCR扩增和测序技术,研究mtDNA D-loop区和mtDNA Cyt b基因的序列特征,利用邻接法构建系统发育树和中介网络关系,以此阐明鹅喉羚南疆亚种的遗传多样性水平和系统进化地位。结果表明:鹅喉羚南疆亚种mtDNA D-loop区序列碱基A、T含量为60.38%,存在23个多态位点,占总位点数的2.90%,其中包括单一多态位点8个,简约性信息位点15个,存在6处插入和缺失,14个个体存在7个单倍型;平均单倍型多样度为0.813,核苷酸多样度为0.00890,平均核苷酸差异度为7.747。mtDNA Cyt b基因序列碱基A、T含量为58.39%,存在5个多态位点,占总位点数的0.46%,其中包括单一多态位点1个,简约性信息位点4个,14个个体存在4个单倍型;平均单倍型多样度为0.714,核苷酸多样度为0.00159,平均核苷酸差异度为1.725。以上数据表明鹅喉羚南疆亚种单倍型多样度远高于核苷酸多样度,具有非常丰富的遗传多样性。系统发育分析表明,鹅喉羚南疆亚种与其他亚种出现了明显的遗传分化,聚类于明显不同的进化支。网络关系分析表明,鹅喉羚具有明显的地理分布特征,研究涉及的鹅喉羚南疆亚种的单倍型大多聚于同一进化支,建议在南疆地区设立自然保护区,加强对鹅喉羚栖息地的生态保护,从而提高其生存适应性。
In order to further clarify the genetic differentiation,genetic diversity and phylogenetic status of Gazella subgutturosa yarkandensis,PCR amplification and sequencing techniques were used to study the sequence characteristics of mtDNA D-loop region and mtDNA Cyt b region of G.subgutturosa in Southern Xinjiang.The genetic diversity and phylogenetic status of the Southern Xinjiang subspecies of goitred gazelle were elucidated by constructing phylogenetic tree and intermediary network relations using the adjacent method.The results showed that:the mtDNA D-loop region of G.s.yarkandensis was rich in bases A and T,with A content of 60.38%.There were 23 polymorphic sites,accounting for 2.90%of the total number of sites,including eight single-type polymorphic sites,15 parsimonious information sites,and six insertions and deletions.The frequency of transformation was much higher than that of transmutation.There were seven haplotypes in 14 individuals.The mean haplotype diversity,nucleotide diversity and mean nucleotide difference were 0.813,0.00890 and 7.747 respectively.The mtDNA Cyt b region of G.s.yarkandensis was rich in bases A and T,with A content of 58.39%.In the region of mtDNA Cyt b,there were five polymorphic sites,accounting for 0.46%of the total number of sites,including one single type polymorphism site and four parsimonious information sites.Four haplotypes were found in 14 individuals.The mean haplotype diversity,nucleotide diversity and mean nucleotide difference were 0.714,0.00159 and 1.725 respectively.The above data indicated that the haplotype diversity of the Southern Xinjiang subspecies was much higher than the nucleotide diversity and had a very rich genetic diversity.Phylogenetic analysis showed that the Southern Xinjiang subspecies of goitelin showed distinct genetic differentiation from other subspecies,showing distinct evolutionary cladistics.The analysis of the network relationship showed that the goitelin had obvious geographical distribution characteristics and most of the haplotypes of the
