摘要
目的观察蒙药给旺-9对D-氨基半乳糖(D-Galactosamine,D-GalN)诱导的急性肝衰竭大鼠保肝、抗炎作用及肝组织形态学改变的影响。方法将72只SPF级雄性SD大鼠,随机法分为空白对照组、模型组、阳性对照美能15 mg·kg^(-1)剂量组、蒙药给旺-9低(0.4 g·kg^(-1))、中(0.8 g·kg^(-1))、高(1.6 g·kg^(-1))剂量组,每天灌胃1次,连续10天。采用D-GalN腹腔注射诱导急性肝衰竭大鼠模型。造模后观察各组大鼠行为学改变,并在造模后48 h取各组大鼠血清和肝组织,采用试剂盒检测血清谷丙转氨酶(Alanine aminotransferase,ALT)、谷草转氨酶(Aspartate aminotransferase,AST),总胆红素(Total bilirubin,T-Bil)、直接胆红素(Direct bilirubin,D-Bil)的活性。ELISA试剂盒检测肝匀浆内毒素(Lipopolysaccharides,LPS)、白介素-4(Interleukin-4,IL-4)、白介素-10(Interleukin-10,IL-10)的含量。血浆分析仪检测凝血酶原时间(Prothrombin time,PT)、活化部分凝血活酶(Activated partial thromboplastin time,APTT)国际标准化比值(International normalized ratio,INR)。肝组织右叶做HE染色,观察组织形态学变化。结果与空白组比较,模型组大鼠血清ALT、AST、T-Bil、D-Bil水平升高(P<0.01),肝匀浆LPS、IL-4、IL-10水平升高(P<0.01),与正常组比较模型组PT、APTT、INR显著升高。差异有统计学意义(P<0.05),与模型组比较阳性对照组、蒙药给旺-9味高、中、低剂量组大鼠PT、APTT、INR无显著降低(P>0.05),显微镜下肝组织出现水肿,炎细胞浸润、坏死等病变。与模型组比较,给旺-9低、中、高剂量组、阳性对照组大鼠血清ALT、T-Bil、D-Bil值显著下降(P<0.05),肝匀浆LPS、IL-4、IL-10含量显著下降(P<0.01),肝组织形态学病变显著改善。结论蒙药给旺-9可保护DGalN诱导的急性肝衰竭大鼠肝功能受损,抗炎并可明显减轻肝脏的病理损害,对肝脏组织起到一定保护作用。
Objective To observe the effects of Gei Wang-9 on the hepatoprotecting,anti-inflammatory and histopathology changes of rats'acute liver disease induced by D-Galactosamine.Methods 72 SPF male SD rats were randomly divided into blank control group,model group,positive control Meineng 15 mg·kg^(-1) dose group,Mongolian medicine Gei Wang-9 low(0.4 g·kg^(-1)),medium(0.8 g·kg^(-1)),high(1.6 g·kg^(-1))dose group,gavage once a day for 10 consecutive days.A rat model of acute liver failure was induced by intraperitoneal injection of D-GalN.After modeling,the behavioral changes of rats in each group were observed,and the serum and liver tissues of rats in each group were collected 48 h after modeling,and the serum ALT,AST,T-Bil,D-Bil,activity were detected by the kit.The ELISA kit detects the content of LPS,IL-4 and IL-10.The plasma analyzer detected PT,APTT,and INR.The right lobe of liver tissue was stained with HE to observe the changes in histomorphology.Results Compared with the blank group,the levels of serum ALT,AST,T-Bil and D-Bil in the model group increased(P<0.01),and the levels of LPS,IL-4 and IL-10 in liver homogenate increased(P<0.01).Compared with the normal group,PT,APTT and INR in the model group increased significantly.The difference was statistically significant(P<0.05).Compared with the model group,PT,APTT and INR in the positive control group and Gei Wang-9 high,medium and low dose groups were not significantly reduced(P>0.05).Edema,inflammatory cell infiltration and necrosis appeared in the liver tissue under the microscope.Compared with the model group,the values of serum ALT,T-Bil and D-Bil in the low,medium and high dose groups and the positive control group of Gei Wang-9 decreased significantly(P<0.05),and the contents of LPS,IL-4 and IL-10 in liver homogenate decreased significantly(P<0.01).The morphological changes of liver were significantly improved.Conclusion Mongolian medicine Gei Wang-9 can protect rats'liver tissue from acute liver disease by improving the hepatoprotecting,antiimflamato
