免疫性血小板减少症患者外周血CD4^(+)CD25^(+)CD127^(low) Treg细胞与炎症因子、趋化因子的相关性研究 被引量:3

摘要 目的:研究免疫性血小板减少症(ITP)患者外周血CD4^(+)CD25^(+)CD127^(low)Treg细胞与炎症因子、趋化因子的相关性。方法:选择2018年1月~2020年12月期间在某院诊断为ITP的120例患者作为研究的ITP组,同期在我院儿科门诊进行健康体检的儿童100例作为对照组,测定外周血中CD4^(+)CD25^(+)CD127^(low)Treg细胞的含量以及血清中炎症因子、趋化因子的含量。采用t检验比较两组间上述指标的差异,采用Pearson检验分析上述指标的相关性。结果:ITP组患者外周血中CD4^(+)CD25^(+)CD127^(low)Treg细胞的含量以及血清中IL-4、IL-5、TGF-β、IL-10的含量明显低于对照组,血清中IFN-γ、TNF-α、IL-2、IL-17A、CCL5、CXCL11、MCP-1的含量明显高于对照组,且CD4^(+)CD25^(+)CD127^(low)Treg细胞的含量与IFN-γ、TNF-α、IL-2、IL-17A、CCL5、CXCL11、MCP-1的含量呈负相关(r=-0.615、-0.688、-0.587、-0.535、-0.493、-0.528、-0.703,P=0.010、0.004、0.013、0.024、0.034、0.025、0.000),与IL-4、IL-5、TGF-β、IL-10的含量呈正相关(r=0.626、0.591、0.652、0.571,P=0.009、0.012、0.007、0.018)。结论:ITP患者外周血CD4^(+)CD25^(+)CD127^(low)Treg细胞减少与促炎因子、趋化因子的增多以及抑炎因子的减少有关,其参与病情的发展变化。 Objective:To study the correlation between peripheral blood CD4^(+)CD25^(+)CD127^(low)Treg cells with inflammatory factors,chemokines in patients with immune thrombocytopenia(ITP).Methods:120 cases of children diagnosed with ITP in a hospital from January 2018 to December 2020 were selected as the ITP group and 100 children who underwent physical examination in the hospital were selected as the control group.The levels of CD4^(+)CD25^(+)CD127^(low)Treg cells in peripheral blood and serum inflammatory cytokines,chemokines were measured.The differences of the above indexes between the two groups were compared by t-test,and the correlation of the above indexes was analyzed by Pearson test.Results:The levels of CD4^(+)-CD25^(+)CD127^(low)Treg cells in peripheral blood and the contents of IL-4,IL-5,TGF-β,IL-10 in serum of ITP group were significantly lower than those of control group,the contents of IFN-γ,TNF-α,IL-2,IL-17A,CCL5,CXCL11 and MCP-1 in serum were significantly higher than those of control group.The levels of CD4^(+)CD25^(+)CD127^(low)Treg were negatively correlated with IFN-γ,TNF-α,IL-2,IL-17A,CCL5,CXCL11 and MCP-1(r=-0.615,-0.688,-0.587,-0.535,-0.493,-0.528,-0.520.528,-0.703,P=0.010,0.004,0.013,0.024,0.034,0.025,0.000),and were positively correlated with IL-4,IL-5,TGF-β,IL-10(r=0.626,0.591,0.652,0.571,P=0.009,0.012,0.007,0.018).Conclusion:The decrease of CD4^(+)CD25^(+)CD127^(low)Treg cells in peripheral blood of ITP patients correlates with the increase of pro-inflammatory cytokines,chemokines and the decrease of anti-inflammatory cytokines,and is involved in the development of the disease.
