摘要
为研究不同产奶量奶山羊机体氧化应激状态变化规律,根据富平县奶山羊平均产奶量,将奶山羊分为高产组(日均产奶量大于2.5 kg)和低产组(日均产奶量小于2.5 kg),分别检测不同组奶山羊的体温、呼吸数、脉搏数以及乳成分差异,同时采集奶山羊血清样品检测机体氧化应激状态(总抗氧化水平、H_(2)O_(2)、NO、MDA含量)差异,采集乳汁样品检测炎症相关因子(IL-1β、IL-6、IL-8、IL-10等)mRNA的表达变化。结果显示:高产奶山羊与低产奶山羊在体温、呼吸数和脉搏数上无显著差异(P>0.05);高产奶山羊与低产奶山羊的乳脂率、乳糖率和总固体率差异显著(P<0.05);与低产奶山羊相比,高产奶山羊血清总抗氧化水平显著降低(P<0.05),氧自由基H_(2)O_(2)水平显著升高(P<0.05);乳体细胞炎症趋化因子CCL5、IL-8的mRNA表达水平显著升高(P<0.05),促炎因子IL-1β、IL-6的mRNA表达水平亦明显升高(P<0.05),抗炎因子IL-10的mRNA表达量明显下降(P<0.0001)。研究表明,高产奶山羊体内氧化和抗氧化系统失衡,氧化应激状态升高,导致机体倾向于炎症状态,抗炎能力下降,增加了高产奶山羊对疾病的易感性。
To investigate the variation of the oxidative stress status in dairy goats with different milk yields,dairy goats in Fuping County were used and divided into the high-yield group(with an average daily milk yield of over 2.5 kg)and the low-yield group(with an average daily milk yield of less than 2.5 kg)based on the average daily milk yield per goat there.Then,the differences in body temperature,respiration rate,pulse rate,and milk composition between the high-yield and low-yield groups were detected.Serum samples of the dairy goats were collected to detect the differences in the oxidative stress state(the total antioxidant level,the H_(2)O_(2),NO,and MDA contents),and milk samples were collected to detect the mRNA expression levels of inflammation-related factors(IL-1β,IL-6,IL-8,IL-10,etc.).The results showed that there were no significant differences in body temperature,respiration rate,and pulse rate between the high-yield and low-yield groups(P>0.05),but there were significant differences in milk fat rate,lactose rate,and total solids rate between the two groups of dairy goats(P<0.05).Compared with the low-yield dairy goats,the serum total antioxidant level of the high-yield dairy goats decreased significantly(P<0.05),but their oxygen free radical H_(2)O_(2) level increased significantly(P<0.05).The mRNA levels of inflammatory chemokines CCL5 and IL-8 in the somatic cells isolated from their milk increased significantly(P<0.05),and the pro-inflammatory factors IL-1βand IL-6 also increased significantly(P<0.05),but the anti-inflammatory factor IL-10 decreased significantly(P<0.0001)in the high-yield group.In summary,the present results indicated that the oxidation and antioxidant systems in high-yield dairy goats were unbalanced,and the oxidative stress increased,which led to the tendency of inflammation and lowered the anti-inflammatory ability,resulting in higher susceptibility to diseases in high-yield dairy goats.
