摘要
利用蚕豆的22个主要农艺性状和8个简单重复序列(Simple sequence repeat,SSR)标记对“第三次全国农作物种质资源普查与收集行动”中收集到的57份江西省蚕豆种质资源进行遗传多样性分析。结果表明:江西省地方蚕豆种质资源具有丰富的遗传多样性,单株分枝数及单株总荚数与蚕豆籽粒的大小呈显著负相关。农艺性状聚类分析将57份种质资源分为两大类,其中第Ⅰ类群单株产量较高,可作为潜在高产优质蚕豆育种材料。8个SSR标记共扩增出65条清晰、可识别的条带,其中多态性条带49条,多态率为75.38%。通过NTSYS聚类分析,在遗传相似性系数为0.743处将57份蚕豆种质资源分为3个类群,这些类群中不同来源的种质相互交错,表明江西省蚕豆资源间的基因交流较充分。综合以上结果,筛选出4份优异的蚕豆资源,分别具有产量高、抗性强、高钙高蛋白质含量等特点,可作为蚕豆新品种的育种材料。研究结果可为蚕豆种质资源的评价利用及蚕豆育种亲本的选择等提供理论基础。
In this study,22 main agronomic characters and eight simple sequence repeat(SSR)markers were used to study and analyze the genetic diversity of 57 broad been germplasm resources collected in the third national investigation and collection of agricultural germplasm resources.The results showed that the local broad bean germplasm resources in Jiangxi province had rich genetic diversity,and the number of branches per plant and total pods were significantly negatively correlated with the size of broad bean seeds.The 57 broad bean germplasm resources were divided into two categories by cluster analysis,the group I had higher yield per plant and could be used for high-yield breeding.A total of 65 clear and identifiable bands were amplified by eight SSR markers,including 49 polymorphic bands,with a polymorphism rate of 75.38%.According to NTSYS cluster analysis,57 broad bean germplasm resources could be divided into three groups at the genetic similarity coefficient of 0.743.The germplasms from different sources in these groups were interlaced,indicating that broad bean resources in Jiangxi were fully exchanged.Based on the above results,four excellent broad bean resources with high yield,strong resistance,high calcium and high protein content were screened and could be used as breeding materials for new varieties of broad bean.The research results can provide theoretical basis for the evaluation and utilization of broad bean germplasm resources and the selection of parents for broad bean breeding.
