81个矮牵牛自交系遗传多样性的分析

摘要 【目的】对81个矮牵牛自交系的亲缘关系进行分析。【方法】采用十六烷基三甲基溴化铵法提取矮牵牛幼叶基因组DNA,利用扩增片段长度多态性对81个矮牵牛自交系的基因组DNA进行酶切、连接、预扩增、选择性扩增。【结果】采用8对引物组合共扩增出24 351条谱带,多态性条带比率为100%,平均每对引物扩增出3 044个多态性位点。81个矮牵牛自交系的相似性系数在0.2443~0.6233之间,平均值0.4338;每个位点的有效等位基因数1.2236,平均多样性指数0.2422;遗传相似性系数较高的是4号和5号,达到0.6233,较低的是9号和92号,为0.2443。【结论】根据遗传相似性系数进行聚类分析,可将81个矮牵牛自交系分为3个类群:91号、92号聚为Ⅰ类(小花型自交系);58号、73号和78号聚为第II类(中花型自交系);其余76个聚为第Ⅲ类(大花型自交系)。第Ⅲ类根据不同色系又可分为8个小类群,其中11个通过自然变异选育的自交系为D类,与其他类群的遗传关系较远。 【Objectives】 The research on relationship of 81 inbred lines provides knowledge supporting for Petunia hybrid breeding and variety right protection.【Methods】 High-quality genomic DNA extracted from young leaves by the CTAB method was used to analyze genetic diversity and genetic similarity by sequence-related amplified polymorphism molecular markers. Separation of amplified fragments was performed on 6% denaturing polyacrylamide gels. The gels were stained with AgNO;, then "0" or "1" matrix was obtained according to electrophoresis results. The number of polymorphic loci, percentage of polymorphic loci, effective number of alleles, and the indices of genetic diversity were estimated by POPGENE version 32. The genetic similarity estimated by NTSYS pc-V. 2.1 was used for unweighted pair group method analysis(UPGMA) to group the Petunia hybrid germplasm. 【Results】 Among 24351 bands detected by 8 elective primer pairs, 24351 bands(100%) were polymorphic. On average, each primer combination amplified 3044 loci and 3044 polymorphic loci. Genetic similarity of 81 Petunia hybrid inbred lines ranged from 0.2443 to 0.6233 with an average of 0.4338. The number of effective alleles for each locus was 1.2236, and the average index of genetic diversity was 0.2422. The highest genetic similarity(0.6233) was found between line #4 and #5, and the lowest(0.2443) was found between #9 and #92. Petunia hybrid germplasm was divided into 3 groups in term of a threshold of genetic similarity of 0.36. Lines #91 and #92, small flower, were placed in the same member group I. Lines #58, #73, and #78, medium flowers with white, light-pink, and pink color were placed in group II. Other 76 lines were placed in group Ⅲ, which have large flowers. They could be further divided into 8 subgroups with different flower colors. 【Conclusions】 Firstly, cluster analysis shows that 81 Petunia hybrida inbred lines share high genetic similarity and low genetic diversity.
