摘要
为了明确蚕豆中酚类物质的提取工艺,评价青海不同品种(系)蚕豆籽粒中酚类物质含量及抗氧化活性差异,文中采用超声波辅助溶剂法提取蚕豆中酚类物质,通过单因素和正交试验优化出最佳提取工艺参数,测定蚕豆提取液中酚类物质含量及其抗氧化活性。结果表明:蚕豆中酚类物质提取的最佳工艺参数为料液比1∶10、超声波功率450 W、提取温度45℃、提取时间25 min,在此工艺参数条件下测得蚕豆中总酚、原花青素、总黄酮、花色苷含量及其清除DPPH·能力分别为1486.24,54.61,580.13,4.67 mg/100 g和31391.36μmol/100 g。30个不同品种(系)蚕豆籽粒中酚类物质含量及清除DPPH·能力差异显著,蚕豆中酚类物质对自由基的清除具有选择性。筛选出陵西1寸、GF3,GF46,GF47和青蚕14号为青海富含酚类物质的主要蚕豆品种,可作为蚕豆健康产品开发利用的原料。
In order to optimize the extraction process of phenolic substances in broad bean,and to evaluate the difference of phenolic content and antioxidant activity in different varieties(lines)in Qinghai,the phenolic substances in faba bean were extracted by ultrasonic assisted solvent method through single factor and orthogonal test.The optimal extraction process parameters were obtained,and the content of phenolic substances in the faba bean extract and its antioxidant activity were determined.The results showed that the optimum process parameters for the extraction of phenolic compounds in faba bean were 1:10 ratio of material to liquid,450 W ultrasonic power,45℃ extraction temperature and 25 min extraction time.The total phenol,proanthocyanidins total flavonoids,anthocyanins and DPPH removal in faba bean were 1486.24,54.61,580.13,4.67 mg/100 g and 31391.36μmol/100 g,respectively.There were significant differences in the content of phenolic compounds and the ability to scavenge DPPH·in the seeds of 30 different varieties(lines).The phenolic substances in faba beans were selective for the removal of free radicals.The varieties Lingxi 1inch,GF3,GF46,GF47 and Qingshen 14 are rich in phenolic substances,and can be used as raw material for the development and utilization of faba bean health products.
