摘要
通过单因素和正交试验优化出超声波辅助溶剂法提取青海蚕豆多酚的工艺条件,并在最佳工艺条件下研究了青海蚕豆不同品种(系)中酚类物质含量及抗氧化活性。结果表明:甲醇体积分数为60%,料液比为1∶15(g:m L),超声波功率为300 W的条件下35℃提取20 min,蚕豆总酚含量达到47.78 mg/100 g;10种不同蚕豆品种(系)的总酚和总黄酮含量差异显著,其中VF11品种总酚含量最高,2005-00品种总酚含量最低,Y3品种总黄酮含量最高,青蚕15号品种总黄酮含量最低;不同品种(系)蚕豆均表现出较好的DPPH·清除力和还原能力,但·OH、O-2·清除率较弱,且品种间差异明显,其中青海13号、青蚕14号、2005-00、E26对DPPH·清除率较高,达到60%以上,Y3的·OH、O-2·清除率及还原能力均高于其他品种,说明蚕豆中的多酚及黄酮类化合物对各个不同自由基的清除有一定的选择性,且黄酮类化合物可能是蚕豆发挥清除羟自由基、氧自由基及还原能力的主要物质。
To optimize ultrasonic wave-assisted extraction of polyphenols from faba bean, the single factor and . orthogonal experiment were used and the optimum conditions were :60 % methanol as extract solvent, solid-liquid ratio 1 : 15 ( g· mL) ,ultrasonic power 300 W,temperature 35 ℃ and extraction time 20 rain. The Phenolic Content of faba bean was 47.78 mg/100 g. The content of total phenolic ,flavonoids were significantly differed among 10 culfivars of faba beans extracts ( P 〈 0.05) , the highest value of total phenolic and flavonoids content was VFll, Y3 and the weakest was 2005-00 and Qingcanl5. All faba beans bad a stronger capability of scavenging DPPH· and ferric re- ducing ability of plasma, but they had a weeker capability of scavenging ·OH, 02- - ,Antioxidant Activities were sig- nificantly in all cultivars faba beans. The capability of scavenging DPPH · in Qinghail3,Qingean 14,2005-00,E26 were over 60% , Y3 showed stronger scavenging ability in · OH, and O2- · The results showed that polyphenols and flavonoids of faba beans had its own specialty on removing the different free radicals. Flavonoids is the main compound of bean scavenging ability of · OH, O2-· and reducing power.
