摘要
【目的】研究角蛋白关联蛋白1-4基因(KRTAP1-4)多态性对陇东绒山羊羊绒性状的影响。【方法】以382只1岁龄陇东绒山羊为研究对象,测定其产绒量、绒层高度和绒纤维的平均直径。采集试羊皮肤、肾脏、脾脏、肝脏、肺脏、心脏组织及血样,采用逆转录聚合酶链反应(RT-PCR)检测KRTAP1-4基因在6个组织中的表达谱;采用聚合酶链反应-单链构象多态性(PCR-SSCP)检测KRTAP1-4基因的多态性,分析3个羊绒表型性状间的Pearson相关性及KRTAP1-4基因型与羊绒性状间的相关性。【结果】RT-PCR发现,KRTAP1-4基因只在陇东绒山羊的皮肤中表达。在陇东绒山羊KRTAP1-4基因上检测到A、B、C、D 4个等位基因和AA、AB、AC、AD、BB 5种基因型。测序结果表明,山羊KRTAP1-4基因上有1个插入/缺失位点(c.223_252insTGCCAACCGATCTCCATCCAGAC-CAGCTGC)、2个同义突变的单核苷酸多态性(c.333T>C和c.573C>T),以及1个Chi序列(c.14-c.21,5′-CCACCAGC-3′)和2个Chi-like序列(c.46-c.53,5′-ACTGGTGG-3′;c.392-c.399,5′-GCACCAGC-3′)。Pearson相关性系数显示,产绒量与平均纤维直径(r=0.324,P<0.001)及绒层高度(r=0.465,P<0.001)呈中等正相关,平均纤维直径与绒层高度呈弱的正相关(r=0.201,P<0.001)。相关性分析结果表明,AA、AB和AC型山羊个体的产绒量和绒层高度极显著低于BB型个体(P<0.01)。【结论】KRTAP1-4基因可以作为产绒量和绒层高度的分子标记用于陇东绒山羊的育种实践中。
【Objective】This study explored the effects of polymorphisms in keratin-associated protein 1-4 gene(KRTAP1-4)on fiber traits in Longdong cashmere goats.【Method】A total of 382 one-year-old Longdong cashmere goats were selected,and the weight,thickness and mean fiber diameter of cashmere were measured.Tissue samples from skin,kidney,spleen,liver,lung and heart as well as blood samples were collected from these goats,and the expression profiles of KRTAP1-4 were detected using reverse transcription-polymerase chain reaction(RT-PCR).PCR-single stranded conformation polymorphism(PCR-SSCP)was used to detect the polymorphisms of KRTAP1-4.The Pearson correlation among the three cashmere phenotypic traits and the association between genotypes of KRTAP1-4 and cashmere traits were also analyzed.【Result】The RT-PCR results revealed that KRTAP1-4 was only expressed in skin of Longdong cashmere goats.Four alleles of A,B,C and D and five genotypes of AA,AB,AC,AD and BB were detected in KRTAP1-4 of Longdong cashmere goats.The sequencing results showed that there were a insertion/deletion(c.223_252insTGCCAACCGATCTCCATCCAGACCAGCTGC),two synonymous single nucleotide polymorphisms(SNPs;c.333T>C and c.573C>T),a Chi sequence(c.14-c.21,5′-CCACCAGC-3′)and two Chi-like sequences(c.46-c.53,5′-ACTGGTGG-3′and c.392-c.399,5′-GCACCAGC-3′)in caprine KRTAP1-4.The Pearson correlation coefficient indicated that cashmere weight had a moderately high positive correlation with mean fiber diameter(r=0.324,P<0.001)and cashmere thickness(r=0.465,P<0.001),while mean fiber diameter had a weak positive correlation with cashmere thickness(r=0.201,P<0.001).The association analysis showed that goats with genotypes AA,AB and AC had lower cashmere weight and cashmere thickness than those with BB(P<0.01).【Conclusion】Caprine KRTAP1-4 can be used as molecular markers for increasing cashmere fiber weight and cashmere thickness in breeding Longdong cashmere goats.
