自噬在丙泊酚保护糖尿病大鼠肾缺血再灌注损伤中的作用 被引量:6

Role of autophagy in propofol-induced protective effects on renal ischemia-reperfusion injury in diabetic rats

摘要 目的研究丙泊酚对糖尿病(DM)大鼠肾缺血再灌注损伤(IRI)的影响及其机制。方法健康清洁雄性SD大鼠,体质量约200~230 g,采用随机数字表法,将其分为4组(n=15):假手术组(sham组)、DM假手术组(DM+sham组)、DM肾缺血再灌注组(DI/R组)、DM肾缺血再灌注+丙泊酚组(DI/R+P组)。采用开腹夹闭双侧肾动脉60 min,再灌注24 h的方法制备肾IRI模型。DI/R+P组在开放肾动脉前给予输注丙泊酚[25 mg/(kg·h)]120 min。于再灌注24 h后收集各组动脉血及肾组织。用HE染色和光镜观察组织病理变化,自动生化分析仪测定各组血清肌酐(Scr)和尿素氮水平(BUN),Western blot法测定肾组织自噬相关蛋白LC3-Ⅱ蛋白的表达。结果与sham组比较,DI/R组和DI/R+P组大鼠血清Scr、BUN水平升高(P<0.05),肾脏组织LC3-Ⅱ蛋白表达水平升高(P<0.05)。与sham组比较,DM+sham组肾脏组织LC3-Ⅱ蛋白表达水平升高(P<0.05)。与DM+sham组比较,DI/R组和DI/R+P组大鼠血清Scr、BUN水平升高(P<0.05),肾脏组织LC3-Ⅱ蛋白表达水平升高(P<0.05)。与DI/R组比较,DI/R+P组大鼠血清Scr和BUN水平降低,LC3-Ⅱ蛋白表达水平降低(均P<0.05)。结论丙泊酚可减轻DM大鼠肾脏IRI,其机制可能与丙泊酚抑制自噬有关。 Objective To explore the effects of propofol on renal ischemia-reperfusion injury(IRI)in diabetic rats and its mechanism.Methods Healthy clean male SD rats,weighing 200-230 g,were randomly divided into four groups(n=15 in each group):sham group,diabetic sham group(DM+sham group),diabetic renal ischemia-reperfusion group(DI/R group),and diabetic renal ischemia-reperfusion+propofol group(DI/R+P group).The renal IRI model was induced by clamping bilateral renal arteries for 60 min and then reperfusion for 24 h.The rats in DI/R+P group were treated with propofol[25 mg/(kg·h)]for 120 min before opening the renal arteries.Arterial blood and renal tissues were collected after reperfusion for 24 h.HE staining and light microscopy were used to observe the pathological changes.An automatic biochemical analyzer was used to determine the contents of Scr and BUN in the serum.Western blotting was used to detect the protein level of LC3-Ⅱ.Results Compared with sham group,the serum levels of Scr,BUN and the protein expression of LC3-Ⅱin renal tissues were increased in DI/R group and DI/R+P group(P<0.05).Compared with sham group,the expression of LC3-Ⅱin renal tissues was increased in DM+sham group(P<0.05).Compared with DM+sham group,the serum levels of Scr and BUN and the expression of LC3-Ⅱin renal tissues were increased in DI/R group and DI/R+P group(P<0.05).Compared with DI/R group,the serum levels of Scr and BUN and the expression of LC3-Ⅱin renal tissues were increased in DI/R+P group(P<0.05).Conclusion Propofol can alleviate the renal ischemia-reperfusion injury in diabetic rats,which may be related to the inhibition of autophagy.
