芝斛方对气阴两虚证小鼠环核苷酸、免疫及代谢水平的影响 被引量:5

Effects of Zhihu formula on cyclic nucleotide,immunity and metabolism in mice with qi-yin deficiency pattern

摘要 目的研究芝斛方对气阴两虚证小鼠的影响,探讨其益气养阴功效,揭示气阴两虚证实质。方法小鼠随机分为空白组、模型组、生脉颗粒组(5 g/kg)、芝斛方低剂量组(1.33 g/kg)、芝斛方高剂量组(2.67 g/kg);采用单日限制饮食、双日负重力竭游泳,并于第16天开始连续灌胃左甲状腺素钠片(150μg/kg)14 d的方式造模;空白组和模型组给予蒸馏水,其余各组给予相应的药液,连续给药30 d,观察小鼠的一般状态,测试小鼠末次力竭游泳时间,检测小鼠血清环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)、三碘甲状腺原氨酸(T3)、四碘甲状腺原氨酸(T4)、免疫球蛋白G(IgG)、免疫球蛋白M(IgM)、肌肉组织乳酸(LA)、乳酸脱氢酶(LDH)、肌酸激酶(CK)、肌糖原(MG)含量和肝脏组织Na^(+)-K^(+)-ATP酶和Ca^(2+)-Mg^(2+)-ATP酶活性。结果与模型组比较,给药芝斛方的各组小鼠精神状态明显改善,实验第20、30天小鼠的体重明显增加(P<0.01);芝斛方低剂量组和芝斛方高剂量组小鼠的力竭游泳时间均明显延长(P<0.01),胸腺指数增大(P<0.01),血清cAMP含量和cAMP/cGMP比值明显降低(P<0.01),肝脏组织Na^(+)-K^(+)-ATP酶和Ca^(2+)-Mg^(2+)-ATP酶活性增强(P<0.01),肌肉组织LA含量降低(P<0.01),肌肉组织LDH和CK活性增强(P<0.01);芝斛方低剂量组小鼠血清T3含量降低(P<0.05),IgM和IgG含量升高(P<0.05)。结论气阴两虚证反应性在机体不同系统有一定的差异,芝斛方可以改善气阴两虚证小鼠的一般状态,通过调节小鼠的环核苷酸水平、免疫和代谢水平发挥益气养阴功效。 Objective To study the effects of Zhihu formula on mice with qi-yin deficiency pattern,explore its effects of nourishing qi and nourishing yin,and understand qi-yin deficiency pattern.Methods Mice were randomly divided into the blank group,model group,Shengmai granule group(5 g/kg),low-dose Zhihu formula group(1.33 g/kg),and high-dose Zhihu formula group(2.67 g/kg).Mice model was established by using restricted diet,exhausted swimming under gravity,and gavage with levothyroxine sodium tablets for 14 consecutive days.The blank group and model group were given distilled water,and the other groups were given corresponding liquid medicine for 30 days.The general state of the mice was observed,and the last exhausted swimming time of the mice was measured.The serum cAMP,cGMP,T3,T4,IgM,IgG contents,the muscle tissue LA,MG,LDH,CK activity and Na^(+)-K^(+)-ATPase and Ca^(2+)-Mg^(2+)-ATPase activity in the liver tissue were measured.Results Compared with the model group,the mentality of the mice in Zhihu formula groups was all significantly improved,and the weight of the mice increased significantly on the 20 th and 30 th days of the experiment(P<0.01).In the low-dose Zhihu formula group and the high-dose Zhihu formula group,the exhaustive swimming time of mice was significantly prolonged(P<0.01),the thymus index increased(P<0.01),and the serum cAMP content and cAMP/cGMP value were significantly reduced(P<0.01),the activity of Na^(+)-K^(+)-ATPase and Ca^(2+)-Mg^(2+)-ATPase in liver tissue was enhanced(P<0.01),muscle tissue LA contents was reduced(P<0.01),but LDH and CK activity was enhanced(P<0.01).In the low-dose Zhihu formula group,serum T3 contents decreased(P<0.05),and IgM and IgG levels increased(P<0.05).Conclusion There are differences in the responsiveness of qi-yin deficiency pattern to different systems.Zhihu formula can improve the general state of mice with qi-yin deficiency pattern,and regulate the level of cyclic nucleotides,immunity,and metabolism in mice to play effects of nourishing qi and nourishing yin.
