二仙汤对卵巢早衰模型大鼠血清炎性因子、相关生长因子及性激素水平的影响 被引量:13

Effects of Erxian Decoction(二仙汤)on Serum Inflammatory Factor,Related Growth Factor and Sex Hormone Levels in Rats with Premature Ovarian Failure

摘要 目的分析二仙汤对卵巢早衰模型大鼠血清炎性因子、相关生长因子的影响,探讨其治疗卵巢早衰的可能作用机制。方法 60只清洁级健康雌性SD大鼠随机分为正常组、模型组、补佳乐组、二仙汤低剂量组、二仙汤中剂量组、二仙汤高剂量组各10只。除正常组外,其余5组均采用腹腔注射顺铂建立化疗性损伤卵巢早衰大鼠模型。正常组、模型组给予0.5 mL生理盐水灌胃,补佳乐组给予0.5 mL补佳乐溶液(0.006 mg/mL)灌胃,二仙汤低剂量组、二仙汤中剂量组、二仙汤高剂量给予0.5 mL二仙汤(0.234、0.468、0.936 g/mL)灌胃,0.5 mL/次,2次/d,连续4周。比较大鼠血清炎性炎性、相关生长因子、性激素水平等变化。结果与正常组比较,模型组大鼠血清肿瘤坏死因子-α(TNF-α)、γ-干扰素(IFN-γ)含量明显升高(P<0.01),卵巢组织血管内皮生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)蛋白表达明显减少(P<0.01),血清FSH、LH含量明显升高,E_(2)含量明显降低(P<0.01)。与模型组比较,补佳乐组、二仙汤组大鼠血清TNF-α、IFN-γ含量明显降低(P<0.05,P<0.01),卵巢组织VEGF、bFGF蛋白表达明显增加(P<0.05,P<0.01),血清卵泡刺激素(FSH)、黄体生成素(LH)含量明显降低,雌二醇(E_(2))含量明显升高(P<0.05,P<0.01)。与二仙汤低剂量组、二仙汤中剂量组比较,二仙汤高剂量组大鼠血清TNF-α、IFN-γ含量明显降低(P<0.05),卵巢组织VEGF、bFGF蛋白表达明显增加(P<0.05),鼠血清FSH、LH含量明显降低,E_(2)含量明显升高(P<0.05)。结论二仙汤有助于改善卵巢早衰模型大鼠性激素水平,促进卵巢功能恢复,可能与拮抗炎性反应、调节相关生长因子表达水平等因素有关。 Objective To analyze the effects of Erxian Decoction(二仙汤) on serum inflammatory factors and related growth factors in rats with premature ovarian failure(POF), and explore its possible mechanism of action in treating POF. Methods Sixty clean healthy female SD rats were randomly divided into normal group, model group, Progynova group, Erxian Decoction low dose group, Erxian Decoction middle dose group and Erxian Decoction high dose group with 10 rats in each. Except for the normal group, the other five groups were induced by intraperitoneal injection of cisplatin to establish the model of premature ovarian failure induced by chemotherapy. Normal group and model group were given 0.5 mL saline, Progynova group was given 0.5 mL Progynova solution(0.006 mg/mL), Erxian Decoction low, middle and high-dose groups were given 0.5 mL Erxian Decoction(0.234 g/mL, 0.468 g/mL, 0.936 g/mL), 0.5 mL/time, twice a day, totally for 4 weeks. The levels of serum inflammatory, related growth factors and sex hormones in rats were compared. Results Compared with those of the normal group, the serum levels of TNF-α and IFN-γ in the model group were significantly increased(P<0.01). The expressions of vascular endothelial growth factor and basic fibroblast growth factor(bFGF) in ovary tissue were significantly decreased(P<0.01). The serum levels of follicle stimulating hormone(FSH), luteinizing hormone(LH) were significantly increased, and the levels of estradiol(E_(2)) were significantly decreased(P<0.01). Compared with those of the model group, the serum levels of TNF-α and IFN-γ in Progynova group and Erxian Decoction groups were significantly lower(P<0.05, P<0.01). The expressions of vascular endothelial growth factor(VEGF) and bFGF in ovarian tissue were significantly increased(P<0.05, P<0.01). The levels of follicle FSH and LH were significantly lower, and the level of E_(2) was significantlyincreased(P<0.05, P<0.01).
