摘要
以‘津韵1号’草莓茎尖生长点为外植体,探究其最适的诱导和分化一体培养基、增殖培养基、生根培养基的激素浓度和配比,建立了该品种组织培养快繁体系。结果表明:茎尖诱导和分化一体培养基最适宜的激素浓度和配比为MS+6-BA 0.8 mg·L^(-1)+NAA 0.2 mg·L^(-1),诱导率为76.7%,分化率为97%,既可省去愈伤转接操作步骤,又获得了较高的分化率;增殖培养基最适宜的激素浓度和配比为MS+6-BA 0.25 mg·L^(-1)+NAA 0.1 mg·L^(-1),其增殖系数为5.67,生根培养基不添加激素为1/2MS+蔗糖15 g·L^(-1)+琼脂7 g·L^(-1),平均生根数达9.74条。
Taked the stemtip as explant from'JIN Yun No.1'strawberry,studing the optimal hormone concentration and ratio required for induction and differentiation integrated medium,proliferation medium,and rooting medium,and the tissue culture and rapid propagation technology system of the'JIN Yun No.1'strawberry was established.The results showed that the most suitable medium for induction and differentiation integrated medium was MS+6-BA 0.8 mg·L^(-1)+NAA 0.2 mg·L^(-1),and the induction rate was 76.7%,the differentiation rate was 97%.This medium was used as an integrated medium for shoot tip induction and differentiation,which not only saved the procedure of callus transfer,shortened the time of shoot tip formation,but also obtained a high differentiation rate.The proliferation coefficient was 5.67 on MS+6-BA 0.25 mg·L^(-1)+NAA 0.1 mg·L^(-1),and the rooting medium was 1/2MS+Sucrose 15 g·L^(-1)+AGAR 7 g·L^(-1).The average number of rooting reached 9.74.
