摘要
为了研究固醇结合原件蛋白1(SREBP1)基因的表达及其参与脂肪酸代谢的调控机制,以及其与肌内脂肪(IMF)含量的关系,本试验选用相同饲养条件下晋南牛12头,屠宰后测定屠宰性状、背最长肌IMF含量、脂肪酸和氨基酸组分及心脏、脾脏、肾脏、肝脏、背最长肌(LP)、皮下脂肪(SF)、腹腔脂肪(AF)、胸腔脂肪(CF)中SREBP1基因相对表达量。根据IMF含量分为高肌内脂肪组(HIF组)和低肌内脂肪组(LIF组),比较2组LP、SF、AF、CF中SREBP1基因相对表达量的差异,并分析其与IMF含量的相关性。结果表明:LIF组胴体重、屠宰率均显著高于HIF组(P<0.05);HIF组IMF含量为12.30%,显著高于LIF组(P<0.05);LIF组的LP剪切力显著高于HIF组(P<0.05)。HIF组的LP中总脂肪酸(TFA)含量为9.698%,显著高于LIF组(P<0.05)。SREBP1基因在所测组织中均有表达,其中在LP中的相对表达量最高(P<0.05),在肾脏、肝脏和脂肪组织中次之;HIF组SREBP1基因在LP中的相对表达量极显著高于LIF组(P<0.01),在AF和CF中的相对表达量极显著低于LIF组(P<0.01);LP中SREBP1基因相对表达量与IMF含量呈极显著正相关(R^(2)=0.987,P<0.01),AF、CF中SREBP1基因相对表达量与IMF含量则呈极显著负相关(AF:R^(2)=-0.713、P<0.01,CF:R^(2)=-0.946、P<0.01)。综上所述,晋南牛SREBP1基因在不同组织中表达量不同,以LP中相对表达量最高;不同IMF含量晋南牛SREBP1基因表达模式不同,且SREBP1基因正向调控LP中IMF沉积,可作为影响晋南牛IMF含量的候选基因。
To explore the expression mechanism of sterol regulatory element binding protein 1(SREBP1)gene of Jinnan cattle and its role in regulating fatty acid metabolism and the correlation with intramuscular fat(IMF),twelve Jinnan cattle were selected and raised under the same feeding and management conditions.Their slaughtering characteristics,as well as IMF content and fatty acid contents and amino acid composition of longissimus dorsi(LP)were measured after slaughter. The cattle were divided into high IMF content group(HIF group)and low IMF content group(LIF group). The SREBP1 gene expression levels in heart,spleen,kidney,liver,LP,subcutaneous fat(SF),chest fat(CF)and abdominal fat(AF)were detected by real time qPCR,and then their correlations with IMF content were analyzed. The shear force,carcass weight and slaughter rate of LIF group were significantly higher than those of HIF group(P<0.05). The IMF content of HIF group(12.30%)was significantly higher than that of LIF group(4.40%)(P<0.05). The total fatty acid(TFA)content in LP of HIF group was 9.698%,while that of LIF group was 4.064%(P<0.05). Compared with other tissues,the highest relative expression level of SREBP1 gene was in LP(P<0.05),which was relatively higher than that in kidney,liver and adipose tissue. The SREBP1 gene relative expression level in LP of HIF group was strongly significantly higher than that of LIF group(P<0.01). On the contrary,the SREBP1 gene relative expression level in AF and CF of HIF group was strongly significantly lower than that of LIF group(P<0.01). The relative expression level of SREBP1 gene in LP and SF were positively correlated with IMF content(R2= 0.987,P<0.01);oppositely,it in the AF and CF were significantly negatively correlated with IMF content(AF:R2=-0.713,P<0.01;CF:R2=-0.946,P<0.01). In conclusion,the expression level of SREBP1 gene in Jinnan cattle is distinct in different tissues,of which the highest expression level is in LP.
