摘要
目的探讨着丝粒蛋白F(CENPF)基因在肝细胞癌中的表达情况及其与患者临床特征、预后的关系,通过基因集富集分析(GSEA)寻找CENPF基因在肝细胞癌中的相关调控通路。方法从癌症基因组图谱(TCGA)数据库中获取369例肝细胞癌组织样本和50例癌旁组织样本CENPF基因表达数据,以及其中240例病例的临床资料和术后随访资料。CENPF基因表达水平与肝细胞癌患者临床特征的相关性采用χ^(2)检验,CENPF基因表达水平对患者预后的影响采用Cox回归分析,通过GSEA寻找高表达CENPF基因的肝细胞癌样本所富集的基因集。结果肝细胞癌组织中CENPF基因表达水平高于癌旁组织(P<0.05)。肝细胞癌组织中CENPF基因表达水平与病理学分级、血清甲胎蛋白(AFP)水平有关(P<0.05)。多因素Cox回归分析结果显示,TNM分期、血管侵犯和CENPF基因表达水平是肝细胞癌患者死亡的独立危险因素(P<0.05)。GSEA显示,CENPF基因高表达的肝细胞癌样本富集了与DNA复制、剪接体、细胞周期、同源重组、泛素介导的蛋白水解、错配修复等相关的基因集。结论肝细胞癌组织中CENPF基因呈高表达,其表达水平与病理学分级、血清甲胎蛋白(AFP)水平有关,是患者预后不良的危险因素。CENPF基因可能是肝细胞癌患者诊断和治疗的潜在靶点。
Objective To explore the expression of CENPF gene in hepatocellular carcinoma and its correlation with clinical features and prognosis.Gene set enrichment analysis(GSEA)was used to find the relevant regulatory pathways of CENPF gene in hepatocellular carcinoma.Methods The CENPF gene expression data of 369 cases of hepatocellular carcinoma tissue samples and 50 cases of adjacent tissue samples were obtained from the Cancer Genome Atlas(TCGA)database,as well as the clinical data and postoperative follow-up data of 240 cases.The correlation between CENPF gene expression level and the clinical characteristics of hepatocellular carcinoma patients was tested by chi-square test.The effect of CENPF gene expression level on the prognosis of patients was analyzed by Cox regression.GSEA was used to find the gene set enriched in hepatocellular carcinoma samples with high expression of CENPF gene.Results The expression level of CENPF gene in hepatocellular carcinoma tissue was higher than that in adjacent tissues(P<0.05).The expression level of CENPF gene in hepatocellular carcinoma tissue was related to pathological grade and serum alpha-fetoprotein(AFP)level(P<0.05).Multivariate Cox regression analysis showed that TNM staging,vascular invasion and CENPF gene expression levels were independent risk factors for death in patients with hepatocellular carcinoma(P<0.05).GSEA showed that hepatocellular carcinoma samples with high CENPF gene expression were enriched in gene sets related to DNA replication,spliceosome,cell cycle,homologous recombination,ubiquitin-mediated proteolysis,and mismatch repair.Conclusions CENPF gene is highly expressed in hepatocellular carcinoma tissues,and its expression level is related to pathological grade and serum alpha-fetoprotein(AFP)level.CENPF gene is a risk factor for poor prognosis of patients.CENPF gene may be a potential target for diagnosis and treatment of patients with hepatocellular carcinoma.
