摘要 目的:探索抑制着丝粒蛋白F(CENP-F)表达对人胶质母细胞瘤细胞系U251增殖的影响。方法:培养U251细胞,转染抑制片段,检测空白对照组(未做任何处理,Blank组),阴性对照组(转入空白质粒组,NC组)及转入抑制片段组(si-CENPF组,根据转入的干扰片段分si-CENPF#1组、si-CENPF#2组、si-CENPF#3组)各组CENP-F m RNA的表达。挑选抑制最明显的片段进行Western-botting进一步检测CENP-F蛋白水平的表达。行MTT检测转染细胞组与空白对照组及阴性对照组的细胞增殖力。结果:干扰片段能抑制U251细胞系CENP-F m RNA水平及蛋白水平的表达,同时能抑制U251细胞的增殖。结论 :抑制CEAFP的表达能降低人胶质母细胞瘤细胞系U251的增殖。 Objective:To investigate the effect of down-regulation of CENP-F by RNAi on proliferation in human glioma cell line U251. Methods: U251 cell line down-regulating CENP-F by RNAi was established. Forty-eight hours after transient transfection, the expression levels of CENP-F were analyzed by q-PCR and Western-blot;the proliferation of cell line U251 was explored by MTT. Results: In group Si- CENPF, the CENP-F mRNA and protein expression was significantly down- regulated (P 〈 0.05), and the proliferation of cell line U251 was inhibited vs blank control group and negative control group. Conclusion: Down- regulation of CENP-F by RNAi significantly inhibits the proliferation of U251 glioma cell line.
