有氧运动和抗阻运动对肥胖大鼠脂肪组织内质网应激及炎症反应的影响 被引量:8

Effects of Aerobic and Resistance Training on Endoplasmic Reticulum Stress and Inflammation of Adipose Tissue of Obese Rats

摘要 目的:探讨8周有氧运动和抗阻运动对肥胖大鼠脂肪组织内质网应激状态及炎症反应的影响。方法:雄性肥胖SD大鼠30只,随机分为3组:安静对照组(C组)、抗阻运动组(RE组)、有氧运动组(AE组),每组10只。有氧运动方式为跑台训练,速度由15 m/min增至25 m/min,训练时间由20 min增至60 min,每周训练5天,共8周。抗阻运动方式为递增负荷爬梯训练,初始负荷为体重的50%,之后不断增加负荷,每3天进行一次,共训练8周。安静对照组不进行任何训练。最后一次训练结束后48 h取大鼠双侧腹股沟脂肪和附睾脂肪,荧光定量PCR和Western Blot检测观察内质网应激相关因子糖调节蛋白78(GRP78)、类PKR的内质网激酶(PERK)、转录激活因子6(ATF6)和肌醇需求激酶-1(IRE1)以及炎症相关因子白介素6(IL-6)和肿瘤坏死因子α(TNFα)的mRNA和蛋白变化。结果:1. RE组和AE组大鼠体重、附睾和腹股沟脂肪重量均显著低于C组(P<0.01,P<0.05)。2. AE组大鼠两种脂肪的GRP78蛋白相对表达量及IRE1蛋白磷酸化水平显著低于C组及RE组(P<0.01,P<0.05);ATF6和PERK蛋白表达量3组间无显著性差异。3. IL-6蛋白在AE组大鼠两种脂肪中的相对表达量显著低于C组和RE组(P<0.01),RE组显著高于C组(P<0.01)。TNFα仅表现为AE组大鼠在腹股沟脂肪中的相对表达量显著低于C组(P<0.05)。结论:8周有氧运动和抗阻运动均可有效减缓肥胖大鼠的体重增长,降低脂肪重量;相较于抗阻运动,有氧运动能更有效地降低脂肪组织GRP78蛋白的相对表达量和IRE1蛋白的磷酸化水平,进而减轻其内质网应激程度和炎症反应。 Objective To explore the effect of an 8-week aerobic or resistance exercise on the endoplasmic reticulum stress(ERS) and inflammation of adipose issues of obese rats. Methods Thirty obese male Sprague-Dawley rats were randomly divided into a control(C) group,an aerobic exercise(AE)group and a resistance exercise(RE) group,each of 10. Rats in AE group performed treadmill exercise 5 days per week,with the speed increasing from 15 m/min to 25 m/min and the duration increasing from 20 min to 60 min for 8 weeks,while those in group RE climbed a vertical ladder with progressively increased weights once per 3 days for 8 weeks,with the initial weight half of their weight.Rats in group C did not exercise. Then their bilateral inguinal fat(in-WAT) and epididymis fat(epWAT) were isolated 48 h after the last exercise. The quantitative real-time PCR was used to detect the mRNA expression of ERS-related genes including the glucose regulated proteins78/immunoglobul in binding protein(GRP78/Bip),PKR-like endoplasmic reticulum kinase(PERK),activating transcription factor-6(ATF6) and inositol requiring enzyme-1(IRE1),as well as inflammation related genes such as interleukin-6(IL-6) and tumor necrosis factor α(TNFα),while Western blotting was employed to detect the changes in their proteins. Results Compared with the C group,a significant decrease was observed in the average body mass and the weight of in-WAT and ep-WAT in the two exercise groups(P<0.01,P<0.05). The relative expression of GRP78 protein and the IRE1 protein phosphorylation in the two WATs of group AE were significantly lower than group C and RE(P<0.01,P<0.05). However,there was no significant difference in the average PERK and ATF6 expression after 8 weeks’ training.The relative expression of IL-6 protein in the two WATs of group AE was significantly lower than group C and RE(P<0.01,P<0.05),with that of group RE significantly higher than group C,while that of TNFα protein of group AE was significantly lower than group C only in the bilateral inguinal fat(P<0.01
