基于Nrf2信号通路的骨骼肌氧化应激作用探讨藏红花素对小鼠力竭运动疲劳的改善作用 被引量:3

摘要 为研究藏红花素对小鼠运动疲劳的改善和骨骼肌氧化应激损伤的保护作用。通过建立小鼠力竭游泳模型,测定小鼠的力竭游泳时间、抗疲劳和抗氧化相关指标以及骨骼肌中相关基因mRNA的相对表达水平。结果表明,与对照组相比,低、中、高剂量藏红花素均极显著延长了小鼠的力竭游泳时间(P<0.01),其血乳酸(BLA)、血尿素氮(BUN)、丙二醛(MDA)含量和肌酸激酶(CK)乳酸脱氢酶(LDH)活性均呈下降趋势,肝糖原(HG)、肌糖原(MG)含量和抗氧化酶活性逐渐升高;另外,与对照组相比,不同剂量的藏红花素均提高了骨骼肌中抗氧化蛋白(HO-1、NQO1)和抗氧化酶(SOD、CAT、GSH-PX)基因mRNA表达水平。可见,藏红花素能够通过降低骨骼肌内氧化应激水平而发挥抗疲劳作用。 To study the improvement of crocin on exercise fatigue and the protective effect of skeletal muscle oxidative stress injury in mice,establishing the exhaustive swimming model of mice,the exhaustive swimming time,antifatigue and antioxidant-related indicators,and the relative expression levels of related gene mRNAs in skeletal muscle were determined.The results showed that,compared with the control group,low,medium and high doses of crocin significantly prolonged the exhaustive swimming time of mice(P<0.01),and their blood lactate(BLA),blood urea nitrogen(BUN),malondialdehyde(MDA)content and activities of creatine kinase(CK)lactate,dehydrogenase(LDH)showed a downward trend,while liver glycogen(HG),muscle glycogen(MG)content and antioxidant enzyme activity gradually increased;Compared with the control group,different doses of crocin increased the mRNA expression levels of antioxidant proteins(HO-1,NQO1)and antioxidant enzymes(SOD,CAT,GSH-PX)genes in skeletal muscle.It was shown that crocin could exert anti-fatigue effects by reducing the level of oxidative stress in skeletal muscle.
