锌指蛋白PRDM16在肥胖大鼠睾丸及睾周脂肪中的相关性研究 被引量:3

摘要 目的探讨肥胖大鼠睾丸及睾周脂肪中锌指蛋白PRDM16的表达及在睾丸中的定位。方法选取20只SD雄性大鼠,随机分成正常组6只和肥胖组14只,其中正常组普通饲料喂养,肥胖组高脂饲料喂养,12周后成功建立肥胖大鼠模型7只。取大鼠睾丸和双侧睾周脂肪组织称重,采用实时荧光定量PCR和Western Blot检测锌指蛋白PRDM16在两组大鼠睾丸及睾周脂肪中的表达,免疫组织化学法检测PRDM16在大鼠睾丸中的定位,同时留取附睾尾部的精液,检测精子浓度和活力。结果肥胖组大鼠体重(420.43±19.65)g、睾周脂肪质量(5.12±1.75)g显著高于正常组大鼠[分别为(330.80±9.76)g、(2.80±0.23)g](P<0.05),肥胖组大鼠睾丸/体重比值(0.003 9±0.000 44)显著低于正常组大鼠(0.004 8±0.000 37)(P<0.05),而睾丸重量组间无统计学差异[正常组(1.58±0.09)g、肥胖组(1.63±0.15)g,P>0.05];免疫组织化学结果显示,锌指蛋白PRDM16主要表达于大鼠睾丸生精细胞胞质中;Western Blot和qPCR结果显示,与正常组大鼠相比,肥胖组大鼠睾丸及睾周脂肪中PRDM16蛋白表达量和mRNA表达量均显著降低(P<0.05);精液分析结果显示,与正常组相比,肥胖组大鼠精子浓度[(36.14±5.58)×106/ml vs.(46.50±8.10)×106/ml]和活力[(40.57±3.31)%vs.(52.00±6.78)%]均显著降低(P<0.05)。结论肥胖大鼠睾丸及睾周脂肪中PRDM16的蛋白和mRNA表达量均显著降低,推测PRDM16可能与肥胖和雄性生殖有关。 Objective:To investigate the expression of zinc finger protein PRDM16 in testicular and epididymal adipose of obese rata,and its localization in the testis.Methods: Twenty SD male rats were randomly divided into 2 groups:6 rats were fed with normal diet in normal group and 14 rats were fed with high-fat diet in obese group.After 12 weeks,7 obese rats were successfully established.Rat testis and bilateral epididymal adipose were taken and weighted.The mRNA and protein expression of PRDM16 of testicular and epididymal adipose in the two groups were detected by quantitative real-time PCR(qPCR)and Western blot.The localization of PRDM16 in the testis tissue wasdetected by immunohistochemistry.At the same time,semen of the epididymis was taken to measure sperm concentration and motility.Results:Body weight[(420.43±19.65)g vs.(330.80±9.76)g]and epididymal fat mass[(5.12±1.75)g vs.(2.80±0.23)g]in obese group were significantly higher than those in normal group(P0.05).The ratio of obese rats testis/body weight[(0.003 9 ± 0.000 44)vs.(0.004 8 ± 0.000 37)]was significantly lower than the ratio of normal rats(P0.05).There was no significant difference in testicular weight between the two groups[(1.58±0.09)g vs.(1.63±0.15)g](P0.05].The immunohistochemical results showed that the zinc finger protein PRDM16 was mainly expressed in the cytoplasm of spermatogenic cells.Western Blot and qPCR results showed that the PRDM16 protein and mRNA expression in the testicular and epididymal adipose of obese rats were significantly decreased compared with the normal rats(P0.05).The semen analysis showed that the sperm concentration [(36.14±5.58)×106/ml vs.(46.5±8.10)×106/ml]and sperm motility [(40.57±3.31)% vs.(52.00±6.78)%]in obese group were significantly decreased compared with the normal group(P 0.05).Conclusions:The protein and mRNA expression of PRDM16 in the obese rats are significantly decreased,and it is suggested that PRDM16 may be relat
