摘要
目的研究金雀异黄素(GEN)对子宫肌瘤模型大鼠雌激素和孕激素水平及子宫组织增殖细胞核抗原(PCNA)、碱性成纤维细胞生长因子(bFGF)蛋白表达的影响。方法用雌、孕激素负荷法诱导构建子宫肌瘤模型大鼠,将成功构建的子宫肌瘤模型大鼠随机分为模型组与实验组,各12只;选择12只大鼠作为对照组。实验组灌胃金雀异黄素20 mg·kg^(-1)·d^(-1),模型组与对照组灌胃等量生理盐水,连续干预15 d。用放射免疫分析法检测血清雌二醇(E2)、孕酮(P)与促卵泡成熟素(FSH)、促黄体生成素(LH)含量;用苏木精-伊红(HE)染色法观察大鼠子宫组织病理特点;测定子宫干重和子宫系数;用免疫组化法检测子宫组织PCNA、bFGF蛋白的表达情况。结果对照组、模型组与实验组大鼠血清E2含量分别为(127.87±31.92),(439.24±76.11),(215.03±35.84)pg·mL^(-1);P含量分别为(0.34±0.05),(1.21±0.14),(0.72±0.06)ng·mL^(-1);FSH含量分别为(14.91±1.02),(15.06±1.72),(14.83±1.14)U·L^(-1);LH含量分别为(5.91±1.28),(19.37±4.86),(11.02±3.93)U·L^(-1);子宫组织PCNA蛋白阳性细胞百分比分别为(24.32±2.84)%,(86.37±4.21)%,(31.16±5.22)%;bFGF蛋白阳性细胞百分比分别为(33.42±2.93)%,(82.47±5.25)%,(48.68±5.46)%。与对照组相比,模型组大鼠血清E2、P、LH水平与子宫干重、子宫系数均明显升高,且实验组显著低于模型组;模型组大鼠子宫组织PCNA、bFGF蛋白阳性细胞百分比均显著高于对照组,且实验组显著低于模型组(均P<0.05)。结论金雀异黄素可降低子宫肌瘤大鼠血清中雌孕激素水平,同时降低子宫组织PCNA、bFGF蛋白表达,抑制肌瘤细胞的增生和血管生成,从而达治疗子宫肌瘤的目的。
Objective To investigate the effects of genistein(GEN)on estrogen and progesterone levels,uterine tissue proliferating cell nuclear antigen(PCNA),and basic fibroblast growth factor(bFGF)protein expression in uterine fibroid model rats.Methods The uterine fibroid model rats were induced by estrogen and progestin loading methods,the successfully constructed uterine fibroids model rats were randomly divided into model group and test group,with 12 rats in each group;and 12 normal rats were selected as control group.Rats in test group were treated with GEN(20 mg·kg^(-1)·d^(-1))by gavage,model group and control group were treated with normal saline by gavage for 15 d.The serum estrogen(E2),progesterone(P),follicle stimulating hormone(FSH)and luteinizing hormone(LH)contents were determined by radioimmunoassay;hematoxylin-eosin(HE)staining was used to observe the pathological characteristics of rat uterus;the uterine dry weight and uterine coefficient were determined;the expression of PCNA and b FGF protein in uterine tissues were detected by immunohistochemistry.Results The serum E2 content in control group,model group and test group were(127.87±31.92),(439.24±76.11),(215.03±35.84)pg·mL^(-1);P content were(0.34±0.05),(1.21±0.14),(0.72±0.06)ng·mL^(-1);FSH content were(14.91±1.02),(15.06±1.72),(14.83±1.14)U·L^(-1);LH content were(5.91±1.28),(19.37±4.86),(11.02±3.93)U·L^(-1);the percentage of PCNA protein-positive cells in uterine tissues were(24.32±2.84)%,(86.37±4.21)%and(31.16±5.22)%,the percentage of b FGF protein-positive cells were(33.42±2.93)%,(82.47±5.25)%and(48.68±5.46)%.Compared with control group,the serum E2,P,LH levels,uterine dry weight,and uterine coefficient in model group were significantly increased,and test group was significantly lower than model group;the percentage of PCNA and b FGF protein-positive cells in the uterine tissues of model group was significantly higher than that of control group,and testl group was significantly lower than that of model group(all P<0.05).Conclusio
